Reactive oxygen species(ROS) along with other radicals are involved in a variety of biological phenomena including mutation, carcinogenesis, degenerative and other diseases, inflammation, aging, and development. The continuous efflux of ROS form any sources result in continuous and accumulative oxidative damage to cellular components. Among these ROS superoxide ion radical(O2-) which can be converted to other ROS is important.As a scavenger of O2- ,Superoxide dismutase (SOD) plays a very important role in cell.In this study, tumor cells,HepG2 and SPC-A-1, were treated with Rhodamine/FITC labeled Cu,Zn-SOD-magnetoliposomes. The fluorescenct discrimination biodistribution of dyes within cells were observed by LCSM, a time dependent uptake of dyes was detected. The affection of external magnet to SOD-magnetoliposome uptake by cells were investigated, resulting in an enhanced uptake with magnet. DHE was employed to investigate the cellular O2-,after incubation with SOD-magnetoliposomes, a lower O2- level was observed after treatment.After intravenous injection of SOD-magnetoliposome, the right kidney was exposured to external magnet (0.4Tesla),left kidney served as control. After 4hrs of induction, kidneys were resected and frozen sections were made. Fluorescenct of the right kidney was obviously stronger than the left one observed by LCSM., indicating the preparation of SOD-magnetoliposomes posses the anticipated targeting properties.
