Varicocele(VC) is an common disease in department of andrology and urology. Its total incidence is 10%-15% in common males. The incidence of varicocele in male patients with primary infertility syndrome is even high to 30%-50%.The recent study indicates the varicocele has been the first reason leading to infertility. Varicocele would be harmful to both of spermary though it is usually occurred in left. The effect of varicocele on generational function is powerful and be paid more and more attentions to. Currently, spermatic varicocele reproductive hormone changes of various different reports, Inconclusive; Varicocele ligation after changes in reproductive hormones ,And the sperm function, what kind of impact, Still rare reports at home and abroad; In adolescence varicocele infertile mechanism of a very difficult, especially for adolescent varicocele spermatogenic function of pathological more difficult to quantify. In short, so far, the mechanism of infertility in patients with varicocele is still unclear, and curative effect is not satisfactory. It is very important to study the influence of varicocelectomy on generational function.Objective: Explore the changes of mass and volume of spermary and serum reproductive hormone in rats model with varicocelectomy. Estimated the function of producing sperm of spermary in rats refer to Testicular Makler Score. Study the change of characters of reproductive hormone and the function of producing sperm of spermary in rats model with varicocelectomy. Then investigate the mechanism of infertility in patients with VC and the curative effect of varicocelectom on infertility syndrome. Find sensitive indicators of reflect spermatognic function of degree, for the clinical diagnosis, treatment and prognosis of reference.Methods: The male SD rats with weight ranged 180-220g and age ranges 6-7 weeks were choosed into trial. All of 40 rats were randomly divided into 3 groups. Control group(SOG): the left renal vein of ten rats were separated but not ligated. 30 rats were used to build Varicocele model by ligate the left renal vein refer to Saypol's method, 4 weeks later, 15 rats were randomly chosen into varicocelectomy group (SMG) , others were distributed into varicocele group(VG). Three groups were tested after they were raised in the same conditions 8 weeks.Venous blood of each of the rats were gathered, the serum was kept in - 20°C condition. Serum FSH, LH and T were measured by ELISA. The weight and volume of spermary were measured. All of spermary were examined under microscope, and measured diameter of convoluted seminiferous tubule, thickness of convoluted seminiferous tubule, layer and mature level of germ cell were graded by Testicular Makler Score. Then to compare and analyze all of the observed factors in three groups.Results: 1. The level of reproductive hormone: FSH leveles were higher in VG and SMG than in SOG (5.63±0.83IU/L vs 4.37±0.59IU/L, 3.15±0.73 IU/L) , there were significantly difference among three groups(P<0.05). LH levels were also significantly higher in VG and SMG than in SOG (9.53±1.45 IU/L vs 7.72±1.57 IU/L, 5.54±1.13 IU/L, P<0.05) .The level levels of T were lower in VG and SMG than in SOG (2.74±0.57ng/ml,2.94±0.65ng/ml,3.21±0.67ng/ml), but there were no significantly difference among three groups (P>0.05).2. The volume and weight of rats testis : Three group of rats testis about the quality si : SOG (1.86±0.15g,1.97±0.24g) SMG (1.17±0.32g,1.65±0.29g) VG(0.78±0.26g,1.26+0.32g,). Three group of rats testis volume side about :SOG (2.32±0.37ml, 2.46±0.41ml) SMG (1.98±0.54ml,2.21±0.52ml) VG (1.24±0.43ml,1.93±0.34ml,)Three group of rats testis side about quality, volume difference was significant (P<0.05)3. The pathology changes: there were shorter diameter of convoluted seminiferous tubule and thicker wall, reducer layer and lower mature level of germ cell in VG were shorter than in SOG and SMG. Testicular Makler Score were significantly lower in VG than in SOG and SMG.Conclusions: The regulation of reproduction is a complicated course influenced by many factors. Varicocele is an important factor were shorter which harms the spermary function by changing the reproductive hormone level and pathology of spermary, but the situation can be improved by varicocelectomy.1. Varicose serum levels of reproductive hormones FSH, LH are higher . Sperma-togenesis TMS scores arelower. The harm to spermary is bilateral in rats model with varicocele, but more severely in left than right.2. Through varicocele ligation Serum FSH, LH can be decreased ,spermatogenic function TMS scores are increased. But conditions are not completely restored to normal levels .3. Levels of FSH, LH and spermatogenic TMS scores are negatively correlated Clinical detection of FSH and LH may indirectly reflect testicular sperm function . Detection of its standard Can Provide reference of the clinical diagnosis, treatment and prognosis.
