Medical scientists both in civilian and military always pay close attention to the intoxication of cholinesterase inhibitors.In the cholinesterase inhibitor poisoning, there are three main reasons to death including respiratory failure,convulsion and circulatory failure. Nowadays, patients can be successful remedied by reasonable therapy consisting of artificial ventilation and benzodiazepines.The failure of therapy is that the patients have appeared circulatory failure,which is a vicious condition because it has no recation of pressor agents.Now,there are few researches about it and no effective method of treatment.It is important that research the characters of this type of circulatory failure in order to find the therapy special methods.The intoxication of cholinesterase inhibitor induced by the inhibition of centracholine esterase,than M-receptors and N-receptors are agitated by the acetylcholine.In the past,the treatment of the poisioning,more attention was put on the muscarinic receptor antagonism,and the nicotinic receptor antagonism was negated.Recent years,it was found that neuronal nicotinic receptor has some effect on the reaction of the cholinesterase inhibitor poisoning.Benthiactzine was designed and synthesized at our institute.Its chemical structural characteristics differ from those of previous categories of anti-choline drugs.Our earlier studies have established that benthiactzine exhibits strong characteristics and has potential therapeutic advantages for future application in the treatment of the intoxication of cholinesterase inhibitor.In our research,we study the therapeutic effects of benthiactzine against circulatory failure induced by different reasons to identify its specificity treatment of circulatory failure. At the same time, we would illuminate the characters of circulatory failure induced by the intoxication of orgnaophosphorus.1 .Effects of benthiactzine on hemodynamic parameters and electrocardiogram in circulatory failure by DDVP rats.We investigate the changes of hemodynamic parameters and electrocardiogram before and after treatment with benthiactzine against the circulation failures induced by the cholinesterase inhibitor DDVP in rats. The results indicate that in anesthetized rats intoxicated with cholinesterase inhibitor DDVP,the circulation failure was characterized by the significant decrease in hemodynamic parameters including SEP,DBP,MBP,HR,+dp/dtmax,-dp/dtmax,Vpm and +dp/dtmax/IP.But LVDP and IP were increased.The electrocardiogram appeared bradycardia,nodal rhythm and A-V block,etc.The therapeutic effects of benthiactzine can rapidly reverse the hemodynamic parameters and electrocardiogram changes withine 1-3min and 3-5min.Then, we can conclude that benthiactzine can antagonize circulation failures induced by cholinesterase inhibitor DDVP.2.Effects of benthiactzine on hemodynamic parameters and electrocardiogram in circulatory failure by parathion rats.We investigate the changes of hemodynamic parameters and electrocardiogram before and after treatment with benthiactzine against the circulation failures induced by the cholinesterase inhibitor parathion in rats. The results indicate that in anesthetized rats intoxicated with cholinesterase inhibitor parathion,the circulation failure was characterized by the significant decreases in hemodynamic parameters including SBP,DBP,MBP,HR,+dp/dtmax,-dp/dtmax,Vpm and +dp/dtmax/IP.But LVDP and IP were increased.The electrocardiogram appeared bradycardia,nodal rhythm,etc.The therapeutic effects of benthiactzine can rapidly reverse the hemodynamic parameters and electrocardiogram changes withine 0.5-3min and 3-5min.Then,we can conclude that benthiactzine can antagonize circulation failures induced by cholinesterase parathion.3.Effects of benthiactzine on hemodynamic parameters and electrocardiogram in hemorrhagic shock rats.We study the effect on hemodynamic parameters before and after treatment with benthiactzine against the hemorrhagic shock in rats.The therapeutic effects of benthiactzine can reverse the blood pressure and cardiac functions withine 5-30min.The electrocardiogram has no change obviously after administration.4.Effects of benthiactzine on hemodynamic parameters and electrocardiogram in the septic shock rats. We investigate the effect on hemodynamic parameters before and after treatment with benthiactzine against the septic shock in rats.The therapeutic effects of benthiactzine can reverse the blood pressure withine 3-25min.Cardiac functions and electrocardiogram have no change obviously after treatment.5.Effects of benthiactzine on hemodynamic parameters and electrocardiogram in cardiogentic shock dogs.On the cardiogentic shock caused by amidoprocain in dogs to observe the effect on hemodynamic parameters and electrocardiogram before and after treatment with benthiactzine.The therapeutic effects of benthiactzine can reverse the hemodynamic parameters and electrocardiogram changes withine 10 min.From these observations,it could be concluded:1 .The characteristics of circulatory failure induced by cholinesterase inhibitor poisoning are that the models cardiac functions are defeated,the pacing and transimission are damaged.2.The therapeutic effects of benthiactzine can rapidly reverse the hemodynamic parameters and electrocardiogram changes withine 0.5-3min and 3-5min.It can antagonize circulation failures induced by cholinesterase inhibitor effectively and immediately.3.Benthiactzine can antagonize the hemorrhagic shock,septic shock and cardiogentic shock moderately.The therapeutic effects of benthiactzine can reverse the blood pressure and cardiac functions withine 5-30min in the hemorrhagic shock and septic shock rats.The electrocardiogram has no change obviously after treatment.The therapeutic effects of benthiactzine can reverse the hemodynamic parameters and electrocardiogram changes withine 10min in cardiogentic shock dogs.Then,we can conclude that benthiactzine can antagonize circulation failures induced by cholinesterase parathion specially.
