Objective To investigate the therapeutic effect of the combinative treatment of Yunke and colloidal chromic phosphate Phosphonium-32 (32p) compared with the monotherapies in rats with adjuvant arthritis.Methods In this study, 50 Sprague-Dawley rats were randomly assigned to five groups with 10 rats in each group: group A was normal control group, the adjuvant arthritis was induced in the rats by a single intradermal injection of 0.1ml normal saline into the subplantar area of the left hind paws, 0.04ml normal saline was injected into the left hind ankle joints on the 10th day after the arthritis was induced, and the corresponding volume of normal saline (0.25ml/kg) was intra-peritoneal injected every other day; group B was AA control group, the adjuvant arthritis was induced in the rats by a single intradermal injection of 0.1ml Complete Freund Adjuvant into the subplantar area of the left hind paws, 0.04ml normal saline was injected into the left hind ankle joints on the 10th day after the arthritis was induced, and the corresponding volume of normal saline (0.25ml/kg) was intra-peritoneal injected every other day; group C was 32p colloid group, the adjuvant arthritis was induced in the rats by a single intradermal injection of 0.1ml Complete Freund Adjuvant into the subplantar area of the left hind paws, 0.02ml (0.02 mCi) colloidal chromic phosphate Phosphonium-32 and 0.02ml normal saline were injected into the left hind ankle joints on the 10th day after the arthritis was induced, and the corresponding volume of normal saline (0.25ml/kg) was intra-peritoneal injected every other day; group D was Yunke group, the adjuvant arthritis was induced in the rats by a single intradermal injection of 0.1ml Complete Freund Adjuvant into the subplantar area of the left hind paws, 0.04ml normal saline was injected into the left hind ankle joints on the 10th day after the arthritis was induced, and the corresponding volume of Yunke (0.25ml/kg) was intra-peritoneal injected every other day; group E was combination group, the adjuvant arthritis was induced in the rats by a single intradermal injection of 0.1ml Complete Freund Adjuvant into the subplantar area of the left hind paws, 0.02ml colloidal chromic phosphate Phosphonium-32 and 0.02ml normal saline were injected into the left hind ankle joints on the 10th day after the arthritis was induced, and the corresponding volume of Yunke (0.25ml/kg) was intra-peritoneal injected every other day. The left to right diameter of the left hind ankle, arthritic index, articular radionuclide imaging, serum levels of tumor necrosis factor (TNF) and interleukin 1β(IL-1β), and the pathological sections of left hind ankle joints were tested at different time points.Results All the rats were studied. The left to right diameter of the left hind ankle at week 4 was smaller for the combination group compared with 32p colloid alone (7.11±0.34 vs 7.57±0.29, p<0.01). The arthritic index in the combination was lower than 32p colloid alone (7.40±0.52 vs 8.40±0.52, p<0.01 for week 4; 3.88±0.64 vs 5.00±0.54, p<0.01 for week 6), but compared with Yunke alone, the combination had higher arthritic index (9.50±0.53 vs 8.90±0.57, p<0.05 for week 2; 7.404±0.52 vs 6.704±0.68, p<0.01 for week 4). Compared with Yunke or 32p colloid alone, the combination had no statistically difference in the value of T/NT in the range of interest. But compared with the AA control group, all the three treatment groups had lower value of T/NT (combination vs AA control was 1.694±0.19 vs 1.994±0.16, p<0.01 for week 4 and 1.51±0.18 vs 1.78±0.24, p<0.01 for week 6; Yunke alone vs AA control was 1.764±0.30 vs 1.99±0.16, p<0.05 for week 4; 32p colloid alone vs AA control was 1.52±0.18 vs 1.78±0.24, p<0.05 for week 6). The combination was more efficacious than 32p colloid alone in decreasing serum levels of TNF (1.616±0.443 vs 1.977±0.306, p<0.05 for week 4) and IL-1β(0.271±0.033 vs 0.308±0.024, p<0.01 for week 4; 0.209±0.023 vs 0.255±0.016, p<0.01 for week 6). In histological findings, the combination showed less synovium proliferation compared with Yunke alone and fewer inflammatory cells infiltration compared with 32p colloid alone.Conclusion The combination of Yunke and 32p colloid had better therapeutic effect in treatment of adjuvant arthritis in rats compared with Yunke or 32p colloid alone.
