Objectives A. Study the influence of three kinds of biomaterials on theproliferation of lung adenocarcinoma cell; B. study the influence of threekinds of biomaterials on the cell cycle of lung adenocarcinoma cell A549.Materials and Methods This research cultivated lung adenocarcinoma cellA549 with three commonly used biomaterials in clinic, which were silkthread mainly made up of silk fibroin, absorbable thread mainly made upof polygcolic Acid and slide-wire mainly made up of Polypropylene. After72 hours' direct cultivation, a wave length of 490nm was chosen to theenzyme-linked immunodetection apparatus to test the optical density ofeach group by MTT assay, and then calculated the relative growth rate ofcells and observed different influence of the biological materials ontheir multiplication in each group. Meanwhile, the apoptosis rate and cellcycle in each group were determined by flow cytometer. And finally,different influences on the cell cycle of cell A549 were evaluated. Thestatistics analysis was done by SPSS13.0.Results A. Different length of a same biomaterial has different effectson the restriction of growth of cell A549—it increases with theincreasing length (P<0.05). When the length reaches 6 centimeters, therestriction of growth of cell does not increase any more (P>0.05). B. Different length of a same biomaterial has different effect on therestriction of growth of cell L929—it increases with the increasinglength (P<0.05). When the length reaches 6 centimeters, the restrictionof growth of cell does not increase any more (P>0.05).C. The same length of different biomaterial has different effect on theproliferation of cell A549 (P<0.05)—the restrain rate of Polypropyleneis the lowest and the silk fibroin next, but the PGA has the strongestrestriction on A549 cell.D. The same length of different biomaterial has different effect on thegrowth of cell L929 (P<0.05)—the restriction rate of Polypropylene isthe lowest and the silk fibroin next, but the PGA has the strongestrestriction on A549 cell.E. After 72 hours' direct cultivation of cell A549 with three different2-centimeter-length biomaterials, the results showed by flow cytometerhad statistics significance (P<0.05).Conclusion A. When the dosage achieved a certain density, all the threebiomaterials can restrain the proliferation of cell A549. The more lengthof the biomaterials is, the more restrain of proliferation of A549 showeda dose-dependent effect.B. The same length of different biomaterial has different effect on theproliferation of cell A549 and L929.In the course of tumor therapy, wecan choose biomaterials by the position and the need.C. The three kinds of biomaterials affect on the cell cycle ofA549. Different biomaterials have different influence...
