Dynamic Changes Of Basic Fibroblast Growth Factor Expression In Subventricular Zone In Rat Brain During Streptococcus Pneumoniae Meningitis

Objective: To investigate the expression of bFGF in Subventricular Zone(SVZ)of rat brain during Streptococcus Pneumoniae Meningitis and the changesafter antibiotic and dexamethasone amethasone used .in order to providerationale for the research of bFGF and neurogenesis in SVZ during StreptococcusPneumoniae Meningitis, and attempt to understand the biologic role of bFGFduring Streptococcus Pneumoniae Meningitis.Methods: To construct the models of Streptococcus Pneumoniae Meningitis andnormal control via inoculated intracisternally with strainâ…¢streptococcuspneumoniae and the same volume of normal saline solution,respectively. Theexpression of bFGF in SVZ was detected by immunohistochemiscal stainingmethods after 24h, 72h and the 6^th day of inoculation.Results:(1) The expression of bFGF is obviously up-regulated in SVZ at 24h post-infection , the expression declined after 72 hours and the expressiondeclined to normal till the 6^th day . In the brain from infected rats , thestrong positive immunoreactivity was detected in the infiltratedinflammatory cells in leptomeninges, subarachnoid cavity and ventriclechoroid plexus.(2) The infected dexamethasone adjunctive therapy group , the expression ofbFGF is obviously up-regulated in SVZ compared with the controldexamethasone adjunctive therapy group and is greater than the infectivegroup treated by antibiotics and the infective group till the 6^th day andthe normal control group till the 6^th day . (3) the control dexamethasone adjunctive therapy group, the expression of bFGF were obviously greaterthan the normal control group or infective group till the 6^th day , and isgreater than the infective group treated by antibiotics. (4) In the controlgroup ,the expression of bFGF in SVZ is greater SVZ at 24h post-injectionthan the 72 hours and the 6^th day, there were no significant differencebetween the 72 hours and the 6^th day.Conclusions:1,The up-regulation of bFGF during Streptococcus Pneumoniae Meningitismaybe the self-protection of organism and this protection is not enough .2,During Streptococcus Pneumoniae Meningitis , dexamethasone adjunctivetherapy can up-regulate the expression of bFGF in SVZ , maybe profit for theenhancement of bFGF to neurogenesis3,Dexamethasone adjunctive therapy can up-regulate the expression of bFGFin SVZ in normal condition .
