Abstract/Summary:
In pharmaceutical analysis, separation and the quanlitative and quantitative analysisof multi-component are tipically studied. But there are some shortcomings in the analysisof the complex system by the traditional chromatography, such as, fussy operation,time-consuming and laborious, and so on. Chemometrics can finish the analysis quicklywithout separating componets entirely. In this thesis, chemometrics methods, Multivariatecalibration and Multivariate resolution are used to resolve overlapping peaks inpharmaceutical analysis by chromatography. Pattern recognition is used to evaluate thequality of Traditional Chinese Medicine. The studies can provid theorytical basis forresearch in medical use and have important significance in broadening the applicationrange of HPLC and in quality control of Traditional Chinese Medicine. This thesisconsists of six chapters. The works of author are as follows:1. The method determins the six components in Compound Bendazol andHydrochlorothiazide Capsules by HPLC is introduced. The Capsules from differentmedical factories are chose to compare their contents of components. The averagerecoveries of the six componets are between 93.5~104.4%. Contents components fromdifferent factories are evident different. The method is simple and economical, so it canbe fit for the routine analysis of the six components in Compound Bendazol andHydrochloro-thiazide Capsules.2. Determination of flavones in flavone hippophaes tablets by HPLC is performed,HPLC with multivariate curve resolution with alternating least squares(MCR-ALS) isapplied in resolution of overlapping peaks of quercetin and unknown. Quantitative resultis satisfactory. This method is simple and rapid.3. Multivariate curve resolution with alternating least squares(MCR-ALS) andheuristic evolving latent projections(HELP) are used to resovle the overlapping peaks ofReyanning capsule. So quanlitative and quantitative analysis of Ferulic acid can berealized.4. Partail least squares (PLS) is applied in the prediction of concentration of thesystem consist of camptothecin, 7-ethyl-10-camptothecin and the system of irinotecan,7-ethyl-10-cam ptothecin and camptothecin(as impurity)from HPLC-DAD.The concentration error of componets were-4.17~8.31%. The result shows the partail least squares(PLS) can predict concentration even if there is interferent in the analytical system. 5. Pattern recognition is applied in the Pueraria thomsonii from different regions.The result is truth and impersonality, avoiding the error made by people. It can providtheoretical basis for identifying the truth and fake, quantity evaluation of TraditionalChinese Medicine.
