| Objective:Hepatic fibrosis which is a diffusion hepatic lesion formed by one or moreetiopathogenisises' s long or repeated actions is an important patho-component element in many liver diseases' s growth process ,whose result ishepatic cirrhosis which hazards the people' s health seriously. Now thecurative effect of Western medicine to Hepatic fibrosis isn't ideal. But themultistrata and multitude and polynucleation and polyrake combinationpharmacology contribution is the characteristic of complex prescriptiontraditional Chinese drug in anti-Hepatic fibrosis, which makes it having theascendancy in preventing and curing Hepatic fibrosis and revealing the wideperspective. The study through observing the complex prescription' sclinical curative effect of sorting Liver and anti- fibrosis in anti- Hepaticfibrosis to evaluate its curative effect and on the base of this to observethe effect the blood serum which contains the drug of complex prescriptionof softing Liver and anti- fibrosis to the generation of HSC and secretionof collagen which are the center link of the genesis and the development ofHepatic fibrosis to approach mechanism of action of it.Method:1. clinical research: 78 patients were randomly divided into two groups. 37cases were in control group which were treated by Dahuang zhechong wan and41 cases in treatment group which were treated by complex prescription ofsofting Liver and anti- fibrosis and were observed the changes of the clinicalsymptoms (such as hypochondriac pain, yellow appearance of the skin and sclerae, abdominal flatulence, bad appetite, bitter taste and dry mouth) and the indexesof the Liver function (such as ALT, AST, ALB, TBA) and the indexes of Hepaticfibrosis(such as HA, P-Ⅲ-P, C-Ⅳ, LN).2. zoopery research: adopting collagenase living organism perfusionmethod to drive the rats 's liver horizontal neuron and cultivate origincells.The blood serum which contains the drug of complex prescription of softingLiver and anti-fibrosis was made by routine method and deactivated topre-emergency. Putting the blood serum to the being cultivanted liverhorizontal neuron (masculine comparison drug was Colchicine) then after 48hours to detect the indexes adopting the MTT to determine the generation ofthe cells and the method of doping 3H-proline to determine the compositionof the collagen.Results:1. clinical research: The complex prescription of softing Liver and anti-fibrosis can improve the patients' clinical symptoms, such as such ashypochondriac pain, yellow appearance of the skin and sclerae, abdominalflatulence, bad appetite, bitter taste and dry mouth, which is better thanthe control group (p＜0.05 or p＜0.01); After curing the ALT is 50.4±34.6u/L, theAST is 38.7±21.3umol/L, the TBA is 15.7±9.9u/L, which are lower than beforecuring (p＜0.01) and are lower obviously than the control' s (they are 80.8±57.4u/L, 55.34±30.2u/L, 25.6±10.36umol/L) (p＜0.01); the indexes ofHepatic fibrosis indicate that the HA is 64.1±49.4ug/L, the P-Ⅲ-P, the LN is63.7±19.6ug/L, the P-Ⅲ-P is 45.7±33. gug/L, C-Ⅳis 66.7±28.2ug/Laftercuring, which are lower than before curing (p＜0.01), and the HA and the PⅢPare lower the control's (they are 95.3±98.7ug/L, 66.7±46.8ug/L)(p＜0.05).2. zoopery research" the research of cell generation which was checkedby the method of MTT indicates that the extinction grade of the complexprescription of sorting Liver and anti-fibrosis is 0.286±0.060, which islower than the control group (it is 0.428±0.077) (p＜0.01) which is used SodiumChloride, and lower than the group (it is 0.348±0.076) (p＜0.05) which isused Colchicine. The result of the composition of the collagen which waschecked by the method of doping 3H-proline indicates that raditation activityof the the complex prescription of softing Liver and anti- fibrosis is 170.54±21.73, which is lower than the control group (it is 252.77±21.92) (p＜0.01),and lower than the group obviously (it is 0.348±0.076) (p＜0.05) which is used Colchicine.Conclusions:1. The complex prescription of softing Liver and anti- fibrosis can improvethe patients' clinical symptoms and the liver function and lowering theindexes of Hepatic fibrosis indicate obviously. It has manifest treatmenteffect in curing Hepatic fibrosis.2. The blood serum which contains the drugs of the complex prescriptionof sorting Liver and anti- fibrosis can depress the generation of HSC andcomposition of the collagen. Which may be one of the mechanism of curingHepatic fibrosis.
