Vascular endothelial progenitor cells(EPCs) are the precursors of endothelial cells(EC). The disturbance of endothelial function principally show as the decrease of vascular relaxing factor (NO) or the loss of biological availability of NO. The production of NO is mainly catalyzed by the endothelial nitric oxide synthase(eNOS). The deficiency of eNOS promote the development of atherosclerosis.Tong-xing Luo(TXL) capsule can promote the synthesis of nitricoxide(NO), and improve the diastolic function lesion of endothelium. So we investigated the effects of TXL on the number and the ability of proliferation,migration and adhesion of EPCs from peripheral blood. NO is an important message of signal pathway in human endothelial cells. We observe the expression of eNOS mRNA and protein by RT-PCR and Western-blot methods.And the mechanism of the effects of eNOS was discussed also. The number and the function of proliferation, migration and adhesion of EPCs were remarkably increased, which was most prominent at the concentration of TXL of 500μg/ml for 72 hours. RT-PCR and western-blot analysis showed the expression of mRNA and protein of eNOS was significantly increased also. It'slikely to be a new mechanism of EPCs for therapy ischemic disease.
