Effect Of The Chinese Herbal Medcine On Plasma Endothelin And Serum Nitric Oxide Of Diabetic Rats

Objective:Through observing the effect of the chinese herbal medicine on plasma Endothelin and serum Nitric Oxide and some other indexes of the diabetic rats, investigate mechanism of action of preventing and curing the earlier diabetic nephropathy on the chinese herbal medicine.Methods:50 Wistar male rats, four-month old, 200±20g, were selected and randomly divided into four groups:the Normal Contrasted Group(NCG),the Model Contrasted Group (MCG),the Ramipri Treated Group(RTG) and the Chinese herbal medicine Treated Group (CTG).MCG, RTG and CTG were not given food for 10 hours, then MCG, RTG and CTG were injected in peritoneal cavity one time by streptozotoc(53mg/kg).72 hours later, the rats whose blood glucose were more than 16.7mmol/1 were selected as the diabetic rats, NCG and MCG were treated with clear water, while RTG were treated with solution of ramipri, CTG were treated with decoctions of the chinese herbal medicine. Four and eight weeks later, their urine and blood and kidney sample were collected in order, observe items changes:(1) microamount Blood Glucose(BG); (2) Renal Weight/Body Weight(RW/BW); (3)24 hours Urine Albumin (24hUAlb); (4)serum Creatinine(Cr); (5) Blood Urea Nitrogen (BUN); (6) plasma Endothelin(ET); (7)serum Nitric Oxide (NO).Results:1. BG, RW/BW, 24hUAlb, Cr, BUN, ET and NO of MCG or CTG or RTG were evidently increased than the ones of NCG(p＜0.01).2. RW/BW, 24hUAlb, Cr, BUN, ET, NO of CTG or RTG were evidently decreased than the ones of MCG(p＜0.01), while BG of CTG were evidently decreased than the one of MCG(p＜0.05).3. BG, RW/BW, 24hUAlb, Cr, BUN, ET of CTG were evidently decreased than the ones of RTG(p＜0.05), while NO of CTG were evidently decreased than the one of RTG(p＜0.01).Conclusions:1. The chinese herbal medicine have the effects of controlling BG and decreasing RW/BW of the diabetic rats.2. The chinese herbal medicine have the effects of decreasing 24hUAlb, Cr and BUN of the diabetic rats.3. The chinese herbal medicine have the effects of decreasing ET and NO of the diabetic rats, degrading glomcrulus hypertransfusion, high filtration and internal pressure, improving hemodynamics state, lessening pathological lesion of glomcrulus.
