Object: To investigate the correlation expressions and protectivemechanisms of Hsp27 and Caspase-9 in brain tissues after transientforebrain ischemia and further intervention of Minocycline inMongoligan gerbils.Methods: (1)Animal Groups: Fifty healthy male Mongolian gerbilswere devided into four groups: Normal Group (n=5); Sham OperationGroup (n=5); Ischemia Reperfusion Group (I/R) (n=20); MinocyclineIntervention Group (MI/R) (n=20); I/R Group and MI/R Group weredevided into four subgroups (each group n=5): 6 hours, 1 day, 3 days, 7days after I/R ingury; (2) Models: 10 minutes'comon carotid arteryocclusion(CAO) of both sides and reperfusion for 6 hours, 1 day, 3days, 7 days in Mongolian gerbils; The Sham Operation Group only givesegregation but no occlusion; peritoneal injection of Minocycline(45mg/Kg,Bid) is given in the MI/R Group;(3) HE Staining;(4)Immunohistochemistry.Results: Experimental results revealed that Hsp27 and Caspase-9were expressed in neurons and gliacytes of the brain tissue in 50 gerbils.There were few expression of Hsp27 and Caspase-9 in neurons andgliacytes both normal control group of 5 gerbils and sham operationgroup of 5 gerbils, and there was no significant difference between the two groups (P>0.05). Compared to normal control group, theexpressions of Hsp27 and Caspase-9 in neurons and gliacytes ofischemia-reperfusion gerbils was singnificanfly increased at the 6thhour, 1st day, 3rd day and 7th day, especially at the 1st day (p<0.05).Compared to IR group, the expression of Caspase-9 in neurons andgliaeytes of gerbils in Minocyeline intervention group were evidentlydecreased at the 6th hour, lst day, 3rd day and 7th day (p<0.05).Conclusions:1 Expressions of Hsp27 and Caspase-9 in neurons and gliaeytes ofgerbils were significantly increased following cerebralischemia-reperfusion, this suggested that the increased expressionsof Hsp27 could have protected the brain tissue from damage.2 A correlated relationship was observed between Hsp27 and Caspase-9during brain I/R injury.3 In Minocycline intervention groups the expression of Caspase-9 werelower than those in the IR group. Minocycline with Capase-9 mighttake on action in protecting brain tissue.
