Obective: Investigated the expression of p21, cyclinD1 and PCNA protein in cholangiocarcinoma and normal bile duct tissues, in order to find out the role in the invasion and metastasis of cholangiocarcinoma and try to clarify the mechanism. Furthermore, this study can provide theoretical and experimental foundation to treat malignant tumor and prevent metastasis and recurrence with interrupting the process of angiogenesis and lymphangiogenesis.Methods: Collected 46 specimens of patients with diseases of extrahepatic bile hospitalized during 1999-2006 in the xiangya hospital. Among them,35 specimens blocks were diagnosed as cholangiocarcinoma pathologically ,another 11 specimens as normal bile tissues from live transplantation. Before the investigation starting , the diagnoses were confirmed by pathologic experts in the hospital. Immunohistochemistry S-P method was used to determine the expression of p21,cyclinD1 and PCNA in extrabepatic cholangiocarcinoma. Image analysis system was adopted to evaluation their grey scale.Results: 1. Expression of P21 protein:(1)the positive expression rates of P21 in cholangicarcinoma was 42.8%.It was significantly lower than normal bile duct tissue which the positive expression rates of P21 was 72.7%(P<0.05).(2)The positive rate of grade I - II group was lower than that of grade III. P21 was clearly assoaiated with metastssis of cholahgicarcinoma.p21 had no relationship with age,sex and the tumor size.(3)P21 protein was inverse correlation with cyclinD1 and PCNA in the cholangiocarcinoma.2. Expression of cyclinD1 protein:(1)the positive expression rates of cyclinD1 in cholangicarcinoma was significantly higher than normal bile duct tissue.(P<0.05)(2)The positive expression rates of cyclinD1 in cholangicarcinoma with grade I - II was lower than that of grade III CyclinD1 was clearly assoaiated with metastssis of cholangicarcinoma. CyclinD1 had no relationship with age,sex and the tumor size.(3)cyclinD1 was inverse correlation with PCNA in the cholangiocarcinoma.3. Expression of PCNA:(1) the positive expression rates of PCNA in cholangicarcinoma was significantly higher than normal bile duct tissue.(P<0.05)(2)The positive expression rates of PCNA in cholangicarcinoma with grade I - II was lower than that of grade III.There were relative between them. PCNA was clearly assoaiated with metastssis of cholangicarcinoma.Conclusion: 1.P21 was lower expressed in the cholangiocarcinoma; cyclinD1 and PCNA was higher expressed in the cholangiocarcinoma.2. p21,cyclinD1 and PCNA have relations with clinic grade of BDN. cyclinD1 and p21 have relations with lymph node metastasis of BDN.3. There was a relationship between cyclinD1 and PCNA. P21 protein was inverse correlation with cyclinD1 and PCNA in the cholangicarcinoma.
