Background and Objectives The restoration of blood flow to ischemic tissues causes addition aldamage, which is termed reperfusion injury. Ischemia reperfusion injury occurrences frequently on emergency surgery ,It influences severely the recovery of function, even induces to disability or fatality. The good blood circulation causes the tissue and cell to obtain the sufficient oxygen and the nutrients , Discharges the metabolite. Each kind of reason creates the blood supplies interrupt, the irrigation quantity reduces, can cause the tissue and the cell anemic damage. Its mechanism is extremely complex, it discovered in recent years that the mechanism was more than a factor to participated in the multi-mechanism damage process, It is generally to think that leucocyte lies between the inflammation which leads to respond the energy metabolism barrier the oxygen free radical the calcium overload has in the process in it the key function. This research is for the purpose of discussing expression and the significance of the calcium release channel(ryanodine receptors-1) after the skeletal muscle ischemia-reperfusion, for expounded it does the preliminary study in the calcium overload function.Materials and Methods Healthy male SD rats were divided randomly into the groups, establishs the rat hind legs to the model of skeletal muscle ischemia-reperfusion injury, utilizes the reverse transcription polymerase chain reaction to examine the expression of RyR1 in the rat skeleton muscle cell and analyzes its relative concentration by using the software, spectrometric assays were performed for blood serum levels of malondialdehyde(MDA), nitric oxide(NO) , superoxide dismutase(SOD) and skeletal muscle myeloperoxidase (MPO)level,The oedema degree was quantified by calculating the wet weight/Dry weight ratio of skeletal muscle,Light microscope observes skeletal muscle structural change.Results (1)the skeletal muscle lacks the blood to pour into again causes RyR1 mRNA to express is pouring into two hours enhancements again, But after reduces obviously(P<0.05), It has statistics significance. (2) skeletal muscle ischemia-reperfusion resulted in a significant increase in the level of MDA NO MPO and wet weight/Dry weight ratio, But decrease in the level of SOD,It has statistics significance(P<0.05).Conclusions (1)This research first examines when SMIRI using the RT-PCR method the RyR1 mRNA expression, It's helpful in further expounds the SMIRI occurrence mechanism.(2)because of many kinds of mechanism participation, the skeletal muscle ischemia-reperfusion injury causes the skeleton muscle cell RyR1 protein expression to be dysfunction, the free radical which in addition in the cell the calcium overload, organizes to lack when blood produces, together creates certain harm to the body skeleton muscular tissue.(3)for sought prevents and controls SMIRI to open a feasible method to lay the preliminary foundation.
