Research On The Expression Of TSP-1 And VEGF That Promote Renal Interstitial Fibrosis Of Rats With Proteinuria-overload

Objectives In order to explore the correlated basis of the early phase of interstitial fibrosis of interstitial fibrosis in the rats model of protein-overload proteinuria,the expression of TSP-1 and VEGF are investigated in the present present study.Methods About thirty rats were adoppted as the model of protein-overload in the BSA group and fifhteen rats in the control group.Plasm VEGF and TSP-1 were detected by ELISA and Western Blotting to investigate the relationship among urine proteinuria,VEGF,TSP-1 and renal interstitial fibrosis.At the same time,the research revestigate the clinical feature as well as pathophysiological characteristics.Results In contrast to Saline group,the quantity of 24-hour proteinuria of BSA group increased significantly at the first weekend,peaked at the fifth weekend,stayed high level until the ninth weekend.VEGF expression started increasing at the first weekend(Saline group19.87±22.7; BSA group57.81±23.8, P<0.05, peaked at the fifth weekend(Saline group 25.0±20.66; BSA group 187.7±49.8, P< 0.01,remained at high level til the ninth weekend(Saline groupl7.4±9.72; BSA group92.3±18.44, P<0.01. TSP-1 did not increase at the first,but with the progress of the disease, increased significantly at the fifth weekend (Saline group0.58±0.10; BSA group 0.77±0.12, P<0.05; and until the ninth weekend(Saline group0.61±0.08; BSAgroup1.03±0.21, P <0.01. TSP-1 expression was significantly correlated with TIL Score,and negatively correlated with VEGF;There was a significant correlation between expression of VEGF and 24-hour urine protein.Conclusion The research reveal three findings as follows:Firstly,TSP-l may be the early marker of interstitial fibrosis in rats with protein-overload proteinuria;secondly,VEGF is related and colocalized to the 24-hours urine protein;Finally,when the balance between VEGF and TSP-1 was changed and the TSP-1 increased,TSP-l would prevent the expression of VEGF and contribute to the progress of rena interstitial fibrosis.
