Objective: To investigate the effects of adenovirus-mediated human manganese superoxide dismutase (MnSOD) gene transfection on the proliferation, apoptosis and survivin protein expression of human lung cancer cell lines.Methods: Fifty multiplicities of infectivity (MOI) of human MnSOD gene were transiently transfected into human lung cancer cell lines A549 cells and its persister A549/DDP cells via adenovirus vector. Proliferation,apoptosis and survivin protein expression of the two cell lines were determined by MTT assay,Western bolting,Flow cytometry and Direct cell-counting. All experimental data were analyzed with the statistical soft package of SPSS 12.0.Results: 24 hours after the MnSOD gene transfection, MTT assay and Direct cell-counting demonstrate a decreased speed of proliferation and a increased speed of inhibition in the two cell lines (P<0.05). Flow cytometry discovered that the apoptosis have a significant enhancement and proportion of the G1-period cells increased (P<0.05). Western blot analysis and Immunocytochemical technique show a high expression of survivin protein in both of the two cell lines. But 24 hours after the 50 MOI MnSOD gene transfection, the expression of survivin protein shows a significant decrease (P<0.05).Conclusions: Adenovirus-mediated MnSOD gene can inhibit the growth and proliferation of A549 cells and A549/DDP cells and induce their apoptosis as well. And it can also decrease the expression of survivin in lung cancer cells. Maybe the mechanism of MnSOD gene inhibiting the growth and proliferation of lung cancer cells is through the ability of inducing apoptosis and decreasing the expression of survivin.
