Objective: To study the expressions of Nuclear factorkappa-B(NF-κB) and monocyte chemoattractant protein-1 (MCP-1), andthe effects of flunarizine,aspirin on the expressions of NF-κB andMCP-1 in forebrain tissue following transient cerebral ischemicreperfusion in gerbils.Methods: A cerebral ischemia-reperfusion model in gerbils wasestablished by clamping both common carotids. Seventy gerbils wererandomly divided into four groups, including normal control group, shamoperation group, ischemia-reperfusion(IR) group, flunarizine interventiongroup and aspirin intervention group. Gerbils were fed withflunarizine(20mg/kg) the day before experiment in flunarizineintervention group, while Gerbils were fed with aspirin(5mg/kg) twice aday for five days in aspirin intervention group. The expressions of NF-κBand MCP-1 in forebrain tissue were detected usingimmunohistochemistry technique at the 6th hours, 1st day, 3rd and 7th dayafter IR. Experimental results were analyzed with the statistic softpackage of Spss 11.5.Results: In normal control group and sham operation group, therewas basic expression of NF-κB in neurocyte,no significant differencewas fotmd between the two groups (P>0.05). Compared with normalcontrol group, the expressions of NF-κB in IR group were markedlyincreased at the 6th hour,1st day, 3rd day and 7th day, especially at the1st day (p<0.05). There was no expression of MCP-1 in normalcontrol group and sham operation group. The expressions of MCP-1 in IRgroup were seen at the 6th hour, 1st day, 3rd day and 7th day, especially atthe 1st day (p<0.05). Compared with IR group, the expressions ofNF-κB and MCP-1 in flunarizine intervention group and aspirinintervention group were markedly decreased at the 6th hour,1st day, 3rdday and 7th day (p<0.05).Conclusions:1. The expression of NF-κB and MCP-1 exist in the forebrain tissuesfollowing transient cerebral ischemic reperfusion in gerbils, which may play an important role in the ischemic reperfusion injury.2. Down-regulation of the expression of the NF-κB and MCP-1 weredetected after intervention with Flunarizine and aspirin iningerbils,which attenuates forebrain ischemic reperfusion injury.
