Objective: To investigate the effects of recombinant human Granulocyte colony Stimulating factor (rhG-CSF) mobilized bone marrow stem cells on treatment of acute myocardial infarction in rats.Method: 120 wistar rats were randomly divided into 2 groups: control group and treatment group. A rat model was made by opening chest and ligating the left anterior descending (LAD) coronary artery. Three hours after LAD ligation , the rats of treatment group were treated with rhG-CSF (10ug/kg/d) for 5 days and those of control group receive saline. All rats were haemospasiaed to detect cTnI 24hours, 48hours and 1week after LAD ligation then hearts were harvested. Infarcted area were quantified by TTC. HE stain and immunohistochemistry were used to detect the histopathological exam and infiltration of CD34~+ monocytes in the infarction zone. Western blot were used to detect VEGF in the infarction zone.Results:(1) The death rate of the rat was lower in treatment group than control group (P<0.05) .(2) 24hours,48hours and 1 week after the duplication of the rat model ,the level of cTnI was lower in treatment group than control group (P<0.05) .(3)The infracted area decreased in treatment group compared with control group(P<0.05) and the pathological damage is slighter.(4) there were many monocytes infiltrating with CD34~+ expression in the infarction zone of the treatment group while not founding in the control group. (4) 24hours,48hours and 1 week after the duplication of the rat model ,the level of VEGF increased in treatment group compared with control group(P<0.01).Conclusions: (1) The rhG-CSF treatment can decrease the level of cTnI obviously, lighten cardiac muscle damage.(2)In rat AMI model, rhG-CSF can mobilize bone marrow cells to migrate to and live, proliferate in infarcted zones and differentiate to cardiomyocyte-like cells and vascular endothelial cells. The rhG-CSF treatment can decrease infarct area, reduce mortality. The treatment can increase the level of VEGF to promote blood vessel production , improve the blood supply of cardiac muscle and heart function.
