Expression Of BMP-7 Actor In The Renal Interstitial Fibrosis Of Obstructive Nephropathy In Rats And Intervention Study

Objective:Rat unilateral ureteral obstruction(UUO)models were used in our studies. We administed Valsartan and Fosinopril to UUO rats.Through HE stain,routine immunohistochemical,we researched the changes of expression of BMP-7 protein and TGF-β1 protein.We hope our studies can make the actions of BMP-7 clearer.Methods:Forty-eight Rats were randomly assigned into unilateral ureteral obstruction (UUO),treatment and sham-operated groups(SOR).Sixteen per group.Treated group rats were gavaged with valsartan and fosinopril daily.Rats were killed at the 3rd,7th,14th,21st days after UUO,treatment and sham-operated.The protein expression of TGF-β1 and BMP-7 in renal was detected by immunohistochemistry at each time point.Results:(1)In comparison with sham-operated group,tubular lesion,interstitial fibrosis in UUO and treatment groups markedly increased.However,the lesion area in kidneys of treatment group significantly decreased compared with that of UUO group(p＜0.05).(2)Renal BMP-7 protein levels progressively decreased during the progression of obstructive nephropathy.Compared with the UUO group,the levels of BMP-7 protein were significantly increased in treatment group(p＜0.05).(3)The levels of TGF-β1 in UUO group were increase remarkably compared with the SOR (p＜0.05)and treatment groups(p＜0.05)and consistently associated with interstitial fibrosis progressed.Conclusions:(1)This research result manifested that the kidney tubular pathological changes were aggravated along with the obstruction time,the expression of kidney TGF-β1 were significantly increased.(2)BMP-7 plays an important role in the onset and progression of renal interstitial fibrosis. There is a counter balance between TGF-β1 and BMP-7,which,to a great extent,influences the renal interstitial fibrosis.(3)ARB and ACEI increases the expression of BMP-7 through blocking the action of AngⅡ, suppressing the expression of TGF-β1.One of the mechanisms of AngⅡreceptor blocker protective actions is to increase the expression of BMP-7.
