Objective:To investigate the mutation p53 gene in peripheral blood lymphocyte and inducted sputum of coke oven workers.The p53 gene mutation in peripheral blood lymphocyte and inducted sputum regard as biological marker of oven worker.Methods:81 coke oven workers in certain steel factory were selected and divided into three groups:oven top,oven side,oven bottom group.35 workers unexposed occupationslly carcinogenicsubstances were selected as control group,who worked in warehouse of certain steel factory.We investigated their general information age,sex,con-length-of-service,time ofcontacting PAHs, tobacco index and alcohol,and the work environment.Blood and urine samples were collected immediately after a shift at the end of a working day from 81 coke oven workers and 35 control workers.We used the ultrasonic nebulizer to induct sputum of coke oven workers and control workers.The Urinary 1-hydroxypyrene(HpU)level was measured using high performance liquid chromatography(HPLC).We detected p53 gene mutation in peripheralblood lymphocyte and in inducted sputum of coke oven workers and control workers by PCR-SSCP.Results:In peripheral blood lymphocyte and inducted sputum there was significant differrence in these two groups:the exposed groups and the control group is respectively 28.4 %and 11.4%,22.4%and 8.1%mutation rate of p53 gene(P<0.05),OR value was 3.073 and 5.775.Tendency analysis increased sifnificantly in the three exposed groups and the control group(x~2=7.92,x~2=9.70,P<0.01)in blood and sputum.Compared to the control (11.4%),the mutation rate of p53 gene in the oven top group(39.3%)was obviously high er in peripheral blood lymphocyte(P<0.01),OR value was 5.015(95%CI:1.383-18.185),highdose exposure is a risk factor,Compared to the control(8.1%),the mutation rate of p53 gene in the oven top group(33.9%)was obviously higher in inducted suptum(P<0.05),O R value was 10.676,(95%CI:2.121-53.748).Whether the exposed groups or the control group,in peripheral blood lymphocyte the p53 gene mutation rate was not signifinant differrence in sputum.We detected peripheral blood lymphocyte and inducted sputum,17 were detected positive in blood while negative in sputum,14 were detected negative in blood while positive in sputum.Through concordance analysis Kappa value was 0.197,we think the concordance relation is weak.Compared to solo-detection blood or sputum(28.40%and 25.93%),the combination detection(43.21%)was signifinant difference(P<0.05).Tendency analysis increased sifnificantly in the three exposed groups and the control group(x~2=11.40,P<0. 01)in the combination detection p53 gene mutation.Compared to the control(14.29%), the combination detection mutation rate of p53 gene in the oven top group(60.71%)and in the oven side group(37.04%)was obviously higher(P<0.01),OR value was 9.273(95 %CI 2.755~31.191),3.529(95%CI 1.034~12.043).Conclusion:Coke oven contacting could result in increased p53 gene mutation rate in eripheral blood lymphocyte and inducted sputum;p53 gene mutation in eripheral blood lymphocyte are not concordance with in inducted sputum because of mechanism of action.The combination detection of p53 gene in eripheral blood lymphocyte and sputum is increasing detection rate,as a biological marker of exposure oven and injury.
