Objects Formaldehyde(FA)and benzene(BZ)are volatile organic compounds,often coexistence at the variety pollution source and important indoor air pollutants caused by decoration and fitment,probably to expose crowd exertive consociation of toxicity effect.There are the characteristics of high concentration in indoor air and serious biological toxicity to bio-body.But there's not any report about the combined effects of the two substances on the reproductive toxicity now.This study is to observe the alone/combined effects of formaldehyde and benzene on filial fetal of genetic toxicity,to take effective steps can control the indoor air pollution and enhance the quality of human living.Method 4 Kunming Female mice were randomly classified into 11 groups:negative control group,positive control group(used to detect the filial generation foetus mice hepatocyte micronucleus)and nine experiment groups.The formaldehyde poisonous dose is(0.8 mg/m~3,1.6 mg/m~3,3.2mg/m~3).benzene dose is(495mg/m~3,990mg/m~3,1980mg/m~3).Female mice in conceive the 5th day inhaled toxic substance silently for10d,2h/d.The mice in the negative group inhaled normal air.Positive control mice were injected with 40mg/kg cyclophosphamide intreperitoneally for continuous 2 days in conceive the 13th day.Female mice were dissected after they were pregnant for 15 days.The Indexes were observed during the experiment or were determined after the experiment.Results 1.There's no significant difference of implantation number and alive fetus weight between the experimental groups and the negative control group.Fetal absorption rate in all experimental groups were higher than the negative group significantly(P<0.01and P<0.05), while alive fetus number decreased significantly(P<0.01 and P<0.05)2.There's significant difference of fetal liver micronucleus rates between the experimental groups and the negative control group as benzene of formaldehyde exposure dose increased(P<0.01).And this indicated the combined effects of formaldehyde and benzene on fetal liver micronucleus rates(P<0.05) 3.Within tested doses,the number of comet cells in the fetal liver cells significantly in-creased as benzene of formaldehyde exposure dose increased(P<0.01).The length of Tail DNA%and Tail moment was significantly in-creased as benzene increased(P<0.01)but high formaldehyde was obvious low in medium experimental groups as formaldehyde increased(P<0.01).Among combined exposure doses,the Tail DNA%and Tail moment was uniformly increased as benzene increased(P<0.01)but high formaldehyde was obvious low in medium experimental groups as formaldehyde increased(P<0.01).There were the combined effect between benzene and formaldehyde with factorial analysis(P<0.01).Conclusion Under this condition,formaldehyde and benzene could affect embryotoxicity and genetics.The toxicity exposed to formaldehyde and benzene simultaneously is greater than that of exposed to formaldehyde and benzene respectively.Benzene and formaldehyde had the combined effects on promoting the response of embryotoxicity and fetal liver cell each other.
