Objective To explore the therapeutic mechanism of Astragalus on psoriasis so as to provide theoretical basis for clinic by detecting the level of bcl-2 in psoriatic lesions and normal skin from the level of organization and cell.Methods 1. The expression of bcl-2 were detected in 30 cases of lesional psoriatic vulgaris skin and in 28 cases of normal skin by using immunohistochemical method.2.The growth of cultured keratinocyte and the influence after adding different concentration astragalus injection were detected by the means of MTT. 3.The ratio of cultured keratinocyte and the influence after adding different concentration astragalus injection were tested with flow cytometry. 4.The expression of bcl-2 were observed in cultured keratinocyte and the influence after adding different concentration with immunocytochemistry.Results 1. There was significant difference of bcl-2 protein expression between lesional psoriatic vulgaris skin and normal skin.2.Keratinocytes were cultured successfully and testified by Immunohistochemistry.3.MTT showed 10μg/ml concentration had no obviously effect on cultured keratinocyte. 100μg/ml concentration 1000μg/ml concentration 10000μg/ml concentration astragalus had inhibited the growth of cultured keratinocytes.4.The results of flow cytometry showed that the keratinocytes of G1 stage had increased along with the increasing of astragalus concentration. Apoptotic peak was observed in the 10000μg/ml concentration astragalus.5.(1) There was significant difference between acromlegalic keratinocytes and normal keratinocytes in each concentration. (2) There was significant difference in the four groups of acromlegalic keratinocytes by analysis of variance. There was no significant difference in the four groups of normal keratinocytes by analysis of variance. (3)The results of LSD test showed that there was significant difference between 0μg/ml concentration and 100μg/ml concentration 1000μg/ml concentration 10000μg/ml concentration. There was significant difference between 100μg/ml concentration and 1000μg/ml concentration 10000μg/ml concentration. There was no significant difference between 1000μg/ml concentration and 10000μg/ml concentration in acromlegalic keratinocytes. There was no significant difference in the four groups of normal keratinocytes by LSD test.Conclusions 1.The phenomenon of apoptosis in lesional psoriatic vulgaris skin maybe have relate to the dereasing of bcl-2 protein.2.Astragalus injection can inhibit the growth of keratinocytes.3.Astragalus injection can decrease cell apoptosis by means of increasing the expression of bcl-2.
