Study On The Expression State Of IASPP/iASPP-SV In Human Breast Cancer And Association With P53

Purpose: To investigate the expressions of iASPP, iASPP-SV mRNA and protein inhuman breast cancer and the significance. Methods: Cultivate 6 kinds of breastcancer cell lines and a kind of normal breast cell line.45 specimens of breast cancertissues and paired adjacent breast tissues were obtained surgically from 45 breastcancer patients. Withβ-action gene as reference, the iASPP, iASPP-SV mRNA wasmonitored in 7 cell lines and 45 specimens of breast cancer tissues, paired adjacentbreast tissues by semiquantitative reverse transcription-polymerase chainreaction(RT-PCR). Then detect the iASPP, iASPP-SV protein by Western Blotexperiments. Examined the expression of P53 with immunohistochemistry. Result:①Verify the expression of iASPP-SV mRNA and iASPP-SV protein by RT-PCR andWestern Blot experiments;②The expression of iASPP, iASPP-SV mRNA of thebreast cancer tissues were significantly higher than that of the paired adjacent breasttissues (P＜0.05), and the expression of iASPP-SV mRNA was directly correlatedwith the expression of iASPP mRNA ;③The increase of the expression of iASPP,iASPP-SV mRNA in the p53 wild-type breast cancer were significantly higher thanthat in the p53 mutant breast cancer (P＜0.05);④The expression of iASPP,iASPP-SV mRNA of the breast cancer tissues were significantly higher than that ofthe paired adjacent breast tissues in the p53 wild-type breast cancer as while as in thep53 mutant breast cancer;⑤The expression of iASPP,iASPP-SV mRNA of thebreast cancer tissues were not correlated with the clinical stage and pathological type of breast cancer. Conclusion:①Verify the expression of iASPP-SV mRNA andiASPP-SV protein in 7 kinds of cell lines and breast cancer tissues;②iASPP,iASPP-SV may be correlated with the procession of breast cancer, the expression ofiASPP-SV mRNA was directly correlated with the expression of iASPP mRNA.③iASPP, iASPP-SV may function through P53 way in breast cancer.④iASPP,iASPP-SV could function through non-P53 way in breast cancer.⑤The expression ofiASPP, iASPP-SV mRNA of the breast cancer tissues were not correlated with theclinical stage and pathological type of breast cancer.
