Study On Extraction, Isolation Of Effective Part In Semen Cassiae And Crataegus Pinnatifida Bge. And Their Lipid-regulating Function

As one of the clinic familiar diseases, hyperlipidemia is one of the chief factors toplace a premium on atherosclerosis and coronary heart disease. Semen cassiae andCrataegus pinatigide Bge. are traditional herbals to treat hyperlipidemia. In the study,the best extraction and isolation process conditions were discussed, which were aboutanthraquinone glucosides from semen cassiae and total triterpene acid from Crataeguspinatigide Bge.. Whether the proportion mixture groups had the better lipid-regulatingeffect for hyperlipidemic rats and the best proportion were also found out. The contentand the main results were summarized as follow:1. The extraction and isolation of anthraquinone glueosides from semen cassiae,total triterpene acid from Crataegus pinatigide BgeThe dried powder of semen cassiae was extracted by ultrasonic. The best processcondition was that: the powder of Semen Cassiae having been filtered by 40 mesh sievewas soaked with 85% ethanol for 10h, then was extracted under 65℃for 25min airproof,and the Cassia seeds- solvent ratio was 1:10.The mass of total anthyraqinone and freeanthraqinone could reach 25.39mg/g and 16.49mg/g. which was proved that theultrasonic extraction method was superior over the traditional decocting method and theethanol refluxing method. The best isolation technics was that the extraction solutioncontaining 0.65% salt concentration, which was 1.59mg/mL(pH 8.03 with 50% methylalcohol),was adsorption by AB-8 macroporous resin at flow rate 3BV/h, then it wasdesorpted with 4BV,70% ethanol at flow rate 2BV/h.0.34~0.60mm of Crataegus Pinnatifida Bge. powder was extracted with 90%ethanol under 80℃by reflux for 2.5h, and the ratio of liquid to solid was 11:1. Afterconfected original concentration 3.5mg/mL,pH7.18 with 50% methyl alcohol, theextraction solution was adsorption by AB-8 macroporous resin at flow rate2.5BV/h,then it was desorpted with 70% ethanol (pH10)at flow rate 1.0~1.5BV/min.2.The effect of extraction of semen cassiae, Crataegus pinatigide Bge and theirprescriptions on serum lipid of hyperlipidema rats80 SPF male mice were selected and randomly divided into 8 groups which were normal group, hyperlipemic group, xuezhikang positive control group(100mg/kg perday), Semen Cassia anthraquinone glucosides group, Crataegus Pinnatifida Bge totaltriterpene acid group, proportion A,B and C which were made up of differentproportions of anthraquinone glucosides in semen cassiae and total triterpene acid inCrataegus pinatigide Bge. (180mg/kg per day). 1 week after administered with medicine,mice were treated with 75% fresh yelkfluid emulsion by abdominal cavity injection.The levels of TC, TG, LDL-C and ApoB in the therapeutic groups except SemenCassia anthraquinone glucosides group were lower than those in the hyperlipidemicmodel group(P<0.05, P<0.01), especially in different proportion mixture groups,whereas there was no difference in HDL-C and ApoAI between all the therapeuticgroups and hyperlipidemic model group(P>0.05). The content of TC and LDL-C inCrataegus Pinnatifida Bge total triterpene acid group decreased(P<0.05). Proportion Bdecreased TC , LDL-C and ApoB(P<0.05). The levels of TC, TG and LDL-C inproportion C group were lower than the hyperlipidemic model group(P<0.05).Proportion A had the best lipid-rgeulating effect,which decreased TC and LDL-Cprominently(P<0.01),and decreased TG and ApoB(P<0.05).The results suggested that anthraquinone glucosides from semen cassiae, totaltriterpene acid(ursolic acid) from Crataegus pinatigide Bge.might inhibithyperlipidemic formation, and mixture groups were more effective than those of itssingle extract of semen cassiae and Crataegus pinatigide Bge.The best proportion wasanthraquinone glucosides of semen cassiae/total triterpene acid of Crataeguspinatigide Bge proportion A.
