Objective: To observe the therapeutic effects, toxicity and quality of lifeof paclitaxel(PTX) based chemotherapy regimen leucovorin(CF),fluorouracil(5-FU), cisplatin(DDP) and paclitaxel based chemotherapyregimen leucovorin,fluorouracil, oxaliplatin(L-OHP) in the treatment ofadvanced gastric cancer.Methods: 48 patients of advanced gastric cancer diagnosed by pathologywere randomly enrolled into this study. Chemotherapy regimen of group Awas comprised of paclitaxel 60mg/m2, iv gtt d1,8,15; leucovorin 100mg/m2, ivgtt d1-5; fluorouracil 375mg/m2,iv gtt d1-5; cisplatin 15mg/m2, iv gtt d1-5.Chemotherapy regimen of group B was comprised of paclitaxel 60mg/m2, ivgtt d1,8,15; leucovorin 100mg/m2, iv gtt d1-5; fluorouracil 375mg/m2, iv gtt d1-5;oxaliplatin 65mg/m2, iv gtt d1-5. The regimen was given per 28 days. Allenrolled patients received at least two cycles of treatment.Results: After two cycles of chemotherapy, in group A completeresponse was achieved in 1 case, partial response in 13 cases. The overallresponse rate of group A was 58.3%. In group B complete response wasachieved in 2 cases, partial response in 13 cases. The overall response rate of group B was 62.5%. The overall response rate had no significant statisticaldifference between group A and group B (P>0.05). Nausea, vomiting in groupA were more serious than those in group B (P<0.05). Peripheral sensitiveneuropathy had higher incidence in group B (P<0.05). There was nosignificant statistical difference of leucopenia between the two groups(P>0.05).Conclusion: Both paclitaxel based chemotherapy regimenleucovorin, fluorouracil, cisplatin and paclitaxel based chemotherapy regimenleucovorin, fluorouracil, oxaliplatin in the treatment of advanced gastriccancer showed good therapeutic effects. But toxic side effects were lighterand life quality was elevated obviously in group B, which was worthy offurther clinical trial.
