Aim:To investigate the effect of the ligand of Peroxisome Proliferator-activated Receper gammar Rosiglitazone on human promyelocytic leukemic cell (HL60) line in vitro and its possible mechanismMethods:Human promyelocytic leukemic HL60 cell line was cultured in vitro. HL60 cell was treated with various concentration of ROZ or ADM.. Cell proliferation was evaluated by MTT assay. The influence of Rosiglitazone on apoptosis of humam leukemic cell was detemined by AO/EB double fluorescence staining and flow cytometry after PI staning and cell cycle was analyzed using flow cytometry .The expression of PPARγand Bcl-2,NF-κB,Bax protein were analyzed by western blot.Results:Rosiglitazone inhibited proliferation of HL60 cells and its IC50 to HL60 cells is 49.59μmol/L. Rosiglitazone inhibited proliferation of HL60 cells at the concentrations excessed 25.0μmol/L . Rosiglitazone at concentration of 12.5μmol/L can not inhibit proliferation of HL60 cells .AO/EB fluorescence stain and PI staining flow cytometry data showed rosiglitazone at the concentrations of 50,100μmol/L augmented the induction apoptosis of HL60 cell . The cell cycle was arrested at G1 stage by rosiglitazone at the concentration of 50,100μmol/L .Western blot results showed that HL-60 cell treated with ROZ reduced NF-κB,bcl-2 proteins expression and increased Bax proteins expression via actirating PPARγprotein in a concentration and time dependent manner .If PPARγobstructed subject GW9662 was used,the action of rosiglitazone would disappear. Conclusion : (1) Rosiglitazone inhibited proliferation of HL60 cells(2) Rosiglitazone promote the apoptosis induction of humam leukmic cell(3).The chemosentization of rosiglitazone might be associated with downregulation expression of NF-κB,Bcl-2 protein and upregulation expression of Bax protion actirating of PPARγ.
