Objective: This paper aimed at studying the effect of antihypertensive factor in bovine colostrums onintracellular calcium concentration ([Ca2+]i) in adult rat ventricular myocytes induced by KCi, and investigatethe possible mechanism of AFBC to the cardiac muscle contraction's inhibitory action.Methods: Take bovine colostrums as raw materials, methods antihypertensive factor was purified byuttrafiltration, ion exchange chromatography, gelatin filtration chromatogrephy. Through duplicates the gelatinfiltration chromatographic analysis many times, causes the end product to be salt-bearing rate reduces greatly,carries on the analysis using the atomic absorption spectrophotometer to the end product Na content. Usingfluorescent properties Fura-2/AM at cultured cardiomyocytes of newborn rats and freshly isolated adult ratventricular myocytes.The cells loaded with Fura-2 was observed by fluorescence spertrophotometer in order tocompare the effcts of AFBC in different concentration on [Ca2+]i induced by KCl.Results:1. Through duplicates the gelatin filtration chromatography many times, causes the end product saltcontent from 54.15%to 14.69%, had achieved the cell experiment may accept level.2. When to load cell by Hank's fluid aerosol (The extracellular calcium density is approximately1.3mmol/L), 50mmol/LKCl could obviously elevate ventricular myocytes [Ca2+]i (To compare with the restingcondition P<0.01). But to load cell by non-calcium buffer solution, D-Hank's and PBS fluid aerosol,50mmol/LKCl could not obviously affects ventricular myocytes [Ca2+]i (To compare with the resting conditionP>0.05).3. In 0.5mg/mL density scope, AFBC to the resting condition extracellular calcium density have notobviously effects (Compares with the control group P>0.05).4. AFBC has the similar function with verapamil, in 0.5mg/mL density scope, could deprss the increaseof ventricular myocytes [Ca2+]i induced by 50mmol/L KCl.Conclusion: This research observed the effcts of AFBC in different concentration on [Ca2+]i induced byKCI from the cell level. In disturbance nerve and body fluid elimination condition compared the effect ofAFBC and the classical calcium ion antagonist compound verapamil on extracellular. Prompt AFBC has thesimilar function with verapamil, the antihypertensive mechanism of AFBC is through the suppression ofintracellular Ca2+, thus suppression cardiac muscle contraction function. But causes the concrete detail whichthis result appears, still needs to be dicussed further.
