Efficient methods including macroreticular resin column chromatography, high speed countercurrent chromatography(HSCCC) and preparative high performance liquid chromatography were established for the purification of cyclosporins. By these methods, two cyclosporins produced by Fusarium solani were purified .Behavior of cyclosporins in electrospray ionization mass spectrometry (ESI-MS) was studied and their fragmentation patterns were established. Electrospray ionization mass analysis of amino acids after intensive acid hydrolysis of cyclosporins was adopted for the analysis of amino acid residues in cyclosporins, especially the unique amino acid in the position 1 of cyclosporins. These efficient methods and other modern spectrometric methods were applied in the structural determination of two first reported cyclosporins produced by Fusarium solani. For these two cyclosporins, one was identified as cyclosporin L, another one has a molecular weight of 1184.1, which is different from reported cyclosporins. Preliminary experimentals showed that this"new"compound could be dehydrated cyclosporin A or dehydrated cyclosporin H, the exact chemical structure studies of this"new"cyclosporin is underway.Immunosuppressive and antifungal activities of this two cyclosporins were tested and the experimental results were discussed.
