Objective:To study the detoxification effects of Cedemexâ…¡on the morphine dependent withdrawal syndromes,and whether its action mechanism is relative to the monoamine neurotransmitters,nitric oxide and nitric oxide synthase.Methods:1.There were four tests to assess the physical dependence of Cedemexâ…¡:(1)The isolated ileum of guenea pig test:in this test,the changes of tensility of the guenea pig ileum was observed after the treatment with Cedemexâ…¡;(2)The hasten test in mice:the withdrawal jumping and weight loss induced by naloxone after the treatment with Cedemexâ…¡was observed in mice;(3)The spontaneous abstinent test in rats:the weight loss of rats was observed while Cedemexâ…¡was withdrawal;(4)The hasten test in rats:the naroxone precipitated withdrawal syndromes was observed to evaluate the physical dependence of Cedemexâ…¡.2. Four tests were also used to explore the anti-morphine physical dependence effects of Cedemexâ…¡:(1)the morphine-dependent guenea pig ileum test was used to observe the contraction of the isolated ileum after the treatment with Cedemexâ…¡; (2)the morphine-dependent spontaneous abstinent test was performed to observe the effect of Cedemexâ…¡on withdrawal jumping and weight loss in mice;(3)in the morphine-dependent spontaneous abstinent test,the weight loss of the abstinence syndrome was observed in rats treated with Cedemexâ…¡;(4)in the morphine-dependent hasten test in rats,the withdrawal syndrome and weight loss were investigated in rats treated with Cedemexâ…¡.The internal monoamine neurotransmitters such as norepinephrine(NE),dopamine(DA)and 5-hydroxytryptamine(5-HT)were tested by high performance liquid chromatography(HPLC)in mice.The changes of nitric oxide and nitric oxide synthase in morphine dependent and abstinent rats were investigated by the nitrate-reductase method.Results:In the study on the physical dependence of Cedemexâ…¡,it was found that the guenea pig ileum incubated with Cedemexâ…¡could not produce withdrawal contraction.The mice continuously administrated with Cedemexâ…¡did not appeare the withdrawal jumping and weight loss in the spontaneous abstinent test or the hasten test,while the rats treated,with Cedemexâ…¡in the hasten test had not the withdrawal syndrome.In the investigation on the detoxification effects of Cedemexâ…¡,the finding showed that Cedemexâ…¡could inhibit the contraction of morphine-dependent guenea pig ileum dose-dependently;and Cedemexâ…¡also inhibited the withdrawal jumping and weight loss in mice,and these effects in the high dose group of Cedemexâ…¡was even better than those of clonidine.Cedemexâ…¡could also decrease the withdrawal syndrome and weight loss in rats,clonidine decreased significantly the withdrawal syndrome but the effect of Cedemexâ…¡in inhibiting weight loss was much better.Cedemexâ…¡could decrease the contents of monoamine neurotransmitters,nitric oxide and nitric oxide synthase in the morphine-abstinent animals.Conclusion:It is showed that Cedemexâ…¡has no physical dependence.Cedemexâ…¡has a marked curative effects on morphine-dependent withdrawal syndromes.It may be the causation of its detoxification effects that Cedemexâ…¡decrease the contents of monoamine neurotransmitters,nitric oxide and nitric oxide synthase in the morphine-abstinent animals.
