Objective:To detect the mRNA expression of two CTAs(NY-SAR-35 and OY-TES-1)in Retinoblastoma(RB)and investigate the possibility of applying these antigens as the target antigens for RB specific immunotherapy.Methods:With reverse-transcription polymerase chain reaction(RT-PCR),the mRNA expression of NY-SAR-35 and OY-TES-1 in 15 cases of RB tissues,12 cases of nontumorous' retina tissues and 22 cases of other ocular region normal tissues(12 cases of blepharo-cutis,5 cases of musculi oculi and 5 cases of conjunctiva)was detected,samples selected randomly from PCR positive products of each CT gene were directly sequenced.The results of expression and related clinical parameters(sex,age,patho-typing,tumorous size,clinical stage) were analyzed with statistic analysis software.Results:Among 15 RB tissues,NY-SAR-35 mRNA were detected in 60% (9/15),and NY-SAR-35 mRNA can not be detected in both nontumorous' region and other ocular region normal tissues;Among 15 RB tissues,OY-TES-1 mRNA were detected in 86.7%(13/15),and among 12 cases of nontumorous'retina tissues and 22 cases of other ocular region normal tissues, OY-TES-1 were detected in 16.7%(2/12)and 36.4%(8/22).The results of DNA sequencing confirmed that the RT-PCR products were true target cDNA. No relationship was found between the expression of this CTA genes and related clinical parameters(P>0.05).Conclusion:1,NY-SAR-35 is expressed specifically in RB2,OY-TES-1 can express among of RB,nontumorous'retina tissues and other normal tissues,and indicated that OY-TES-1 just is tumor associated antigen not CTA.
