Objective:To investigate gene diagnosis for micrometastasis in lymph nodes in patients with stomach cancer and the feasibility of mucin 1(MUC1)mRNA and cytokeratin 19(CK19)mRNA as molecular marker to detect micrometastasis of gastric cancer.To investigate the relationship between the clinicopathological of tumor depth, size,location,histio-classification and the lymph node metastasis of gastric cancer.The aim of this study were to investigate the pattern of lymph node metastasis(LNM)/ micrometastasis(MM)in the gastric cancer and its clinical significance.Methods:52 patients who underwent curative gastrectomy and lymph node dissection for gastric cancer were analyzed retrospectively.Expression of MUC1 mRNA and CK19 mRNA was detected in 462 lymph nodes taken from 52 patients with stomach cancer.38 lymph nodes from 4 patients with gastric benign diseasesas controls using reverse transcriptase chain reaction(RT-PCR).Results:A total of 1176 lymph nodes taken from 52 patients with average of 22.62 nodes percase had been assessed by clinicopathological hema-toxylineosin(H&E)staining and reverse transcriptase- polymerase chain reaction (RT-PCR).hematoxylin-eosin(H&E)staining expression was detected in 310 lymph nodes.462 lymph nodes had been assessed by RT-PCR and 223 lymph nodes micrometastasis.In these 462 lymph nodes from stomach cancer patients CK19mRNA expression was detected in 180 lymph nodes(38.96%)and MUC1mRNA expression was detected in 136 lymph nodes(29.44%)by RT-PCR.Neither CK19 no MUC1 expression was observed in all the 38 lymph nodes in the control group.Conclusion:The results suggest that the detection of both MUC1 and CK19 mRNA might helpful to diagnose of micrometastasis for stomach cancer and assessed the incidence of micrometastases of lymph nodes in patients with gastric cancer.The tumor depth and tumor histio-classification and tumor location were main factors to influence the LNM rate.
