Objective: Rat mode1s of nonalcoholic fatty liver disease(NAFLD) were established by giving fat-rich diets for our study ,and prepare to approach the aspect of :(1)To study the mechanism of oxidative stress , Lipid peroxidation and Cytochrome P4501A1 in NAFLD.(2)Relationship between Activation of tumor necrosis factor alpha(TNF-a)and nuclear factor kappa B (NF-kB)in NAFLD.(3)The intervention Vitamin E and Sodium selenite on CYP1A1 and correlation factor in NAFLD.Methods :40 SD rats were randomly divided into 5 groups. The normal control group,model group , VE group, Se group and VE+Se group which were respectively fed with normal diets, fat-rich diets and interfering diets. All animals were sacrificed at the end of the 5th week.(1)The liver pathology was observed under the light microscope. (2)Superoxide dismutase (SOD),malondialdehyde (MDA) in serum and liver were determined by biochemistry analysis .(3)The expressions of tumor nec-rosis factor alpha( TNF-a) and nuclear factor kappa B (NF-kB) proteins in heap-tocytes were examined by immunohistochemistry. (4) The expression levels of Cytochrome P4501A1 mRNA in liver tissue were measured by RT-PCR.Result(1)The degree of hepatic steatosis in liver of model group rats were serious, MDA in serum and liver were hightened while SOD lessened, The expressions of NF-kB and TNF-a proteins and levels of Cytochrome P4501A1 mRNA in liver tissue were increased significantly.All difference between two groups cited above are markly in statistics .(2)Compare to the model group the capability of antioxygen in all interfereing group were increased significantly.Appearance in lessened of MDA in serum and liver while SOD hightened , expressions of NF-kB and TNF-a proteins lessened and hepatic steatosis in liver less serious. The levels of Cytochrome P4501A1 mRNA in liver tissue were lessened in VE+Se group. The change of administration of both Vit-amin E and Sodium selenite were more significantly than those of vitamin E and Sodium selenite alone.Conclusions (1) Cytochrome P4501A1 Gene regulation , Lipid peroxidation, activate of TNF-a and NF-kB all related in NAFLD.(2) The activation of TNF-a , NF-kB to promote the progress of NAFLD and related in the regulation of Cytochrome P4501A1.(3)Vitamin E and Sodium selenitecan protect the fat-rich diets rats as well as protection measure of antioxygen can prevention and cure the Lipid peroxidation and activation of TNF-a , NF-kB.The function of administration of both Vit-amin E and Sodium selenite were more sig-nificantly than those of vitamin E and Sodium selenite alone.
