Objectives: To investigate the effects of quercetin on B7 related protein-1(B7RP-1) expression of human tubular epithelial cells (TECs)cultured in vitro .Through this study we aquire the elementary effects that quercetin exert on cell mediated immunity in kidney.Methods: Human TECs were incubated with different concentration quercetin in different time. Cell proliferation was assessed by MTT colorimetric assay. Expression of B7 related protein-1 mRNA was analyzed by reverse transcription polymerase chain reaction(RT-PCR) respectively. Cell immunnoforescence(IF) was performed to detect the experession of B7RP-1 on TECs membrane.Results:1. The effect of quercetin on proliferation of TECs:During 5-15umol/L concentration,quercetin could significantly inhibit TECs proliferation in a higher dose(P<0.05) ,but there was no deffrentia in a lower dose compared with conrtol group (P>0.05) .2. The effect of different dose quercetin on B7RP-1 expression of TECs induced by TNF-α: the expression of B7RP-1 of tubular epithelial cells induced by TNF-αsignificantly increased with different concentration quercetin ,compared with positive conrtol and normal group (p<0.05).3. The effect of the same dose quercetin on expression of B7RP-1 of TECs induced by TNF-αin defferent time points:the expression of B7RP-1 of tubular epithelial cells induced by TNF-αsignificantly increased with the same concentration of quercetin in time-dependent manner between 12 and 48 hour,and there was a linear correlation with r=0.921,P<0.05.4. The effect of different dose quercetin on B7RP-1mRNA expression of TECs induced by TNF-α:the expression of B7RP-1mRNA of tubular epithelial cells induced by TNF-αbecome increased by different concentration of quercetin ,compared with positive conrtol and normal group (p<0.05).This mean quercetin can stimulate tubular epithelial cells to express B7RP-1 on gene level.5. The effect of the same dose quercetin on expression of B7RP-1 of tubular epithelial cells induced by TNF-αin defferent time points:the expression of B7RP-1mRNA of tubular epithelial cells induced by TNF-αbecome increased by the same concentration of quercetin in time-dependent manner,and there was a linear correlation:between 6 and 12 hour, the stastics is r=0.951, P<0.05 and between 24 and 60 hour,the stastics is r=-0.625, P<0.05.Conclusion: quercetin could not inhibit tubular epithelial cells proliferation with a lower dose , but this effect can be realized with a higher dose. Quercetin possibly could up regulate the expression of B7RP-1 on protein and gene level. Quercetin maybe influence the cell mediated immunity to realize the protection effect on kidney.
