Background and objective:Hepatic fibrosis is the result of all kinds of damage factors effect continually onliver, and it will at last result in cirrhosis. The activation of hepatic satellite cells(HSC)is the key stage. With the development of the study on hepatic fibrosis, the faction ofthe cellular adhesion molecule(CAM) is paid more and more attention. During theHepatic fibrosis, the main function of the CAM is to mediate the interaction of allkinds of hepatic cells and extracellular matrix(ECM). Besides, the signal proteinsregulated by integrins and intercellular adhesion molecule-1 (ICAM-1) also take partin the signal way after growth factor receptor's activation. Therefore, CAM can be thenew target site used to treat hepatic fibrosis. At present, researches about therelationship between integrin and hepatic fibrosis are more than E-cadherin. Intracellpart of E-cadherin is connected with cytoskeletal protein, which may take responsiblefor contract of HSC. So it is reasonable that E-cadherins participate the formation ofhepatic fibrosis. While the traditional Chinese medicine, Resveratrol, is graduallypaid more attention for it's abundant pharmacological action. And it is hopeful to be anew drug to anti-fibrosis. This article approach the mechanism of Resveratrol'santi-fibrosis with CAM and illuminate the expression of integrin-β1 and E-cadherinduring different period of hepatic fibrosis with interference of Resveratrol.Methods:100 SD rats, male, randomly divided into model group and normal control. Modelgroup was still divided into little,middle,large dose group. Made rat hepatic fibrosiswith CCl4 hypodermic injection, given different dose of Resveratrol to intragastricadministration. Kill rats at 4,8,12 weeks after making model. Perform RT-PCR withhepatic tissues. Quantitate expression of integrin-β1 and E-cadherin mRNA,pathological section to judge the degree of hepatic fibrosis, PerformImmunohistochemistry to detection expression of integrin-β1 and E-cadherin inhepatic tissues.Result: Pathological sections show liver tissue of all rats of model group have differentdegree of hepatic fibrosis, which confirmed making models successfully. Expressionof integrin-β1 and E-cadherin in hepatic tissues are with one accord. Expression ofnormal control is lower than model control (p<0.01). Expression of early interventionlittle dose,middle dose group and early intervention control have no statisticdifference (p>0.05). Expression of early intervention large dose group is lower thanearly intervention middle dose group(p<0.01). Expression of later intervention littledose,middle dose group and later intervention control have no statistic difference(p>0.05). Later intervention large dose group lower than later intervention middledose group (p<0.05). Later intervention large dose group and early intervention largedose group no statistic difference(p>0.05). Immunohistochemistry shows percentagesof positive cells of early intervention middle dose group and later intervention middledose group are higher than other groups, while model control,early interventioncontrol and later intervention control lower than other groups.Conclusion:1 Expression of integrin-β1 and E-cadherin in hepatic tissues exist in both normal andpathological liver tissue, but it is higher in pathological liver tissue.2 Expression of integrin-β1 and E-cadherin in hepatic fibrosis are with one accord,while expression of E-cadrin are lower than integrin-β1.3 Integrin-β1 and E-cadherin have intimate relationship with hepatic fibrosis.4 The traditional Chinese medicine, Resveratrol, has obvious effect of anti-fibrosisduring experiment of hepatic fibrosis rats at a dose of 200mg/kg.
