Objective: To investigate the effects of bFGF on PPAR-γ2 in the adipogenic differentiation of human MSCs(hMSCs).Methods: hMSCs were obtained from the whole marrow stromal cells and were cultured till the 4th generation. The cells were divided into 6 groups, then bFGF 4ng/mL or adipose revulsant (Rosiglitazone 5μmol/L+ Insulin 10μg/mL) were added in proliferation phase or differentiation phase of these groups. The mRNA of PPAR-γ2 and Glucose transporter 4(GLUT4) were detected by RT-PCR on one day before inducement or the 3rd day of inducement. Oil Red O staining and Transmission Electron Microscope (TEM) were operated at the 28th day of inducement. The experiment datas were analyzed by statistics.Result: There was no PPAR-γ2 expression in no inducement group, while PPAR-γ2 was found expressed in adipose inducement group. Even PPAR-γ2 was expressed before inducement in the condition of bFGF. Oil Red O staining and TEM showed there were many lipids droplets in differentiation cells..Conclusion: bFGF can motivate the expression of PPAR-γ2 in the adipogenic differentiation of hMSCs.
