| Objectives: Human adrenocortical carcinoma is a very exceptional but more malignant tumor, whose incidence is about 1∶1000000 and etiopathogenisis is not clear. About 2/3 patients who were diagnosed adrenocortical carcinoma could be found the local infiltration and metabasis. So, it had a poor treatment and prognosis. Insulin-like growth factor system (IGFs) is a kind of very important polypeptide growth hormone. They have some relationship with the primary adrenocortical tumorigenesis and can modulate its growth in recent study. Through our study we expected to approach objective as fellow: 1. To evaluate the method of human adrenocortical carcinoma SW-13 cell lines in culture and the biological character of SW-13 cell lines. 2. To evaluate the effect of insulin like growth factor and anti-insulin like growth factor receptor-I on the growth and apoptosis of human adrenocortical carcinoma SW-13 cell lines. To inquire into the possible mechanism of these study. 3. To evaluate if we can treat adrenocortical carcinoma by IGFs as a new strategy of treatment. Materials and methods: 1. SA-â…¡A axenic cell cultured machine by purified implement factory of Suzhou; Napco5410 cell cultured box; Ht2 enzyme-marked implement of Biocell; E-600 optical microscope of Nikon; H-600 perspective electron microscope of Hitache; HMIAS-2000 high clarity colorful analytic system of photograph; Powder L-15 medium, MTT, DMSO, trypsin by Sigma company; Fetal bovine serum by Sijiqing company of Hangzhou; SW-13 cell lines from cell lines storehouse of Chinese scientific institute in Shanghai. Recombinant human IGF-â… , Recombinant human IGF-â…¡, anti-insulin like growth factor receptor-I from R&D company; TUNEL test box from Roche company. 2. SW-13 cell lines were cultured in vitro, we observed the shape and structure with optical and electronical microscope, the growth curve of SW-13 cell lines was obtained by means of MTT. 3. The growth diversification curves of insulin like growth factor and anti-insulin like growth factor receptor-I were obtained by means of MTT. To compare SW-13 cultured in insulin like growth factor of different concentrations mixed with anti-insulin like growth factor receptor-I and single used insulin like growth factor or anti-insulin like growth factor receptor-I and contrasted group by the numerical value obtained by means of MTT. 4. SW-13 cells were cultured with Insulin like growth factor or anti-Insulin like growth factor receptor-I or Insulin like growth factor mixed with anti-Insulin like growth factor receptor-I, and SW-13 cells were cultured without Insulin like growth factor or anti-Insulin like growth factor receptor-I was the contrasted group. Apoptosis of SW-13 cells was tested by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL). We observed the modality of cells by microscope.Results: 1. SW-13 cell should be cultured in 37℃, CO2-free and alkalescent conditions. They would stick to the cliff in 6-8 hours, and after 2 days, they would grow logarithmicly. 2. Insulin like growth factor could significantly promote proliferation of SW-13 cell lines, and their function were correlated with their concentrations (P<0.05). Anti-insulin like growth factor receptor-I could inhibit the effect of insulin like growth factor, and they could also inhibit proliferation of SW-13 cell lines (P<0.05). 3. There were significant difference between the rate of apoptosis of each group(P<0.001). The rate of apoptosis of the cells cultured with insulin like growth factor was the lowest, and the rate of apoptosis of the cells cultured with anti-Insulin like growth factor receptor-I was the highest, The rate of apoptosis of the cells cultured with insulin like growth factor mixed with anti-Insulin like growth factor receptor-I and without Insulin like growth factor or anti-Insulin like growth factor receptor-I were between them, the former was higher than the latter. We also observed significant change in the modality of cells cultured with anti-Insulin like growth factor receptor-I.Conclusions: 1. SW-13 cell lines was a kind of stable and applied cell lines, it would be used in extensive domain of research. 2. Insulin like growth factor could promote the growth of human adrenocortical carcinoma SW-13 cell by the insulin like growth factor receptor-I. Anti-insulin like growth factor receptor-I could inhibit the effect of insulin like growth factor, it maybe significant in the treatment of adrenocortical carcinoma. 3. Insulin like growth factor could promote the growth of human adrenocortical carcinoma SW-13 cell by the insulin like growth factor receptor-I, this effect respected to its effect of restraining cells apoptosis. Anti-insulin like growth factor receptor-I could inhibit the effect of insulin like growth factor, it maybe significant in the treatment of adrenocortical carcinoma. |
