The rats were divided into two groups by random: the sham - operated control group and vascular dementia group, permanent occlusion of bilateral common carotid arterie was used to establish the VD model in rats.â‘¡These rats were tested in Morris Water Maze after 6 weeks, and sacrified after sured their ability of learning and memory were lowerd.â‘¢Immunohistochemistry was applied to observe the changes of Glast expression in the hippocampus in a rat models of the VD.â‘£Hybridization in situ and semiquantitative RT-PCR was applied to observe the changes of Glast mRNA expression in the hippocampus in a rat models of the VD..Resultsâ‘ Morris Water Maze: The oriention test showed that the average escape latency of vascular dementia rats was significantly longer than those sham - operated control groups and the space exploring test showed that the lingering time in platform area of vascular dementia rats is shorter than that of the sham - operated control groups after 6weeks when the animal model were operated.â‘¡Immunohistochemistry: Glast expression significantly lowered in VD rats compared to sham- operated control group.â‘¢Hybridization in situ and RT-PCR: Compared to sham- operated control group, Glast mRNA expression in VD rats showed no change by hybridization in situ and RT-PCR;Conclusionâ‘ The kind of rat model reproduced by permanent occlusion of bilateral commoncarotid arteries of rats can result vascular dementia.â‘¡The changes of expression of Glast in the hippocampus in a rat model of vascular dementia were mainly on protein level and no Mrna level. | | Keywords/Search Tags: | vascular dementia, rat, hippocampus, Glast, immunohistochemistry, RT-PCR, hybridization in situ | PDF Full Text Request | Related items |
