Obiective: Manufacture of the anti-tumor bio-derived bone materials which have the characters of slow release anti tumor drug and repair the bone defect after surgery.Study on the characters of slow release anti tumor drug in vitro experimental.Then grafting this material in the rat model with spinal tumor,observating its ability to decrease the local recurrence of the lesion and repair the bone defect after tumor resection. Methods: (1)Sticks of pig cancellous bone were prepared into deantigenicity bio-derived bone materials bydefatting,deproteinisation,and then composed with gelatin and CDDP.Used this material to observe the character of the control release in vitro,measured the concentration of CDDP in the leachate and tumor inhibitory rate.(2)SHZ-88 mammary cancer cell of the rat was subcutaneous injected into female new born rats,form subcutaneous lump 12 days.Then tumor segments was planted into the lumbar vertebral of the female thymectomize rats,to made animal model of spinal tumor.(3)Thirty rats with spinal tumor was devide into 2 groups radom,15 rats either.Group1 was treatment group,which treated with tumor resection+ anti-tumor bio-derived bone materials grafting;Group2 was control group,which treated with tumor resection + deantigenicity bio-derived bone materials grafting.X-ray films were taken two weeks after the surgery,then the involved vertebral bodies were performed pathology examination to observe the local recurrent rate and count the number of tumor cell in the pathological sections.Results: (1)Deantigenicity bio-derived bone material appears white,porous,which has the structure of the nature cancellous bone. Anti-tumor bio-derived bone material appears yellow ,all of the pores were filled with the compound of gelatin and CDDP .In vitro release experiment,CDDP from anti-tumor bio-derived bone materials appears bursing release at 1-3day,and matain stable release in following 10 days,and the tumor inhibitory rate was higher than 50% in 2 weeks. The local concentration of CDDP was about 100μg/ml,higher than the blood drug level injection intraperitoneal.(2)The involved vertebral bodies was observed soft tissue mass,15 days after the surgery ,encapsuled parts of the vertebral body.Bone defect can be see in the sagittal plane,tumor tissue appears gray-white in color and infiltration into the vertebral body.(3)There were 3 rats proved to have the local recurrence and 9 rats in control group by pathological sections,15 days after grafting surgery. The recurrent rate of the treatment group and control group were 20%,60% respectively ,P<0.05.The recurrent rate of the treatment group was lower than the control group obversely.Chose four parts of the pathology sections,count the cell number by Image por plus software.The average cell number of the treatment group was 313.8±1.7,and 565.6±2.8 in control group, P<0.05.The cell number in the treatment group was lower than the control group obversely.Conclusion: (1)Deantigenicity bio-derived bone material was a perfect scaffold materal to load the control drug release system and repair bone defect. Anti-tumor bio-derived bone material has good character of control release ,which can inhibitate the tumor cell proliferation in vitro.(2)Spinal tumor model in rat can simulate the maligent tumor character,which has great signification in the study the character of the Metastatic Spinal Neoplasms,and comparisons of the curative effects of different therapies.(3)Anti-tumor bio-derived bone material can extermination local tumor foci after rescetion surgery by slowly releasing CDDP,and decrease the local recurrence.
