Objective:The aim of the study is to explore the role of PAF in the human sperm acrosome reaction and mechanism of that and the relationaship between PAF and calcium ionophore A23187 on human sperm acrosome reaction.Another aim of the study is to investigate the difference of acrosome reaction rate in fertile and infertile male and to determine the realationship between sperm motility and acrosome reaction in human.Methods:Semen were collected from 32 fertile men aged 27~40 and 19 infertile men aged 27~43.Acrosomal labeling was carried out using fluorescein-labeled pisum sativum agglutinin(FITC-PSA).Human spermatozoa were previously dealed with PureceptionTMgradient centrifugation technique and incubated in medium for 4 hours for capacitation.And then the spermatozoa were treated with an exogenous mixture of PAF(final concentration 10-7mol/L) or/and calcium ionophore A23187(final concentration lmg/ml)in spermwashing medium for 15 minutes for inducing acrosome reaction.The percentages of acrosome reaction of spermatozoa in blank contrast group, A23187-induced group,PAF-induced group,A23187+PAF-induced group were compared and assessed.Result(s):The percentages of acrosome reaction of spermatozoa show significant difference between PAF-induced group and blank contrast group (P<0.01).The percentages of acrosome reaction of spermatozoa show significant difference between A23187-induced group and blank contrast group (P<0.01).The percentages of acrosome reaction of spermatozoa were significantly higher in A23187-induced group than in PAF-induced group (P<0.01).The percentages of acrosome reaction of spermatozoa were higher in A23187+PAF-induced group than in A23187-induced group and PAF-induced group,but the difference was not statistically significant(P>0.05).The percentages of acrosome reaction of spermatozoa in fertile group were higher than in infertile group(P<0.01).The spermatozoa motility were positively correlated with the percentages of acrosome reaction of spermatozoa.Conclusion:PAF and A23187 significantly increased the AR.But the effect of PAF on AR was less than the effect of A23187.AR in fertile men were higher than in infertile men.The spermatozoa motility were positively correlated with the percentages of acrosome reaction of spermatozoa.It is considered that acrosome reaction test can provide scientific basis for evaluating human sperm quality in feitilization.But it is necessary to further study and clarify mechanism of PAE...
