| Smilax glabra was investigated on chemical composition in order to find natural compounds withinhibition against tyrosinase and antioxidant activity, five compounds were isolated from Smilax glabra;The content of resveratrol from Smilax glabra was quantitated by RP-HPLC; The activity of antioxidantwas quantitated by DPPH·method; The total flavonoids of the concentrated extract in Smilax glabra wasquantitated by spectrophotometri method.There were a lot of reports on chemical composition and their pharmacologic action from Smilaxglabra, but studies on inhibition against tyrosinase and antioxidant activitys were not reported. Theinhibition against tyrosinase activity of Smilax glabra and other drugs were compared, the result suggestedSmilax glabra has the highest activity.The cutting of Smilax glabra was extracted by 60% ethanol in ultrasonic wave, the concentratedextract was isolated by silica gel column chromatography and eluted by chloroform and methanol in theway of gradient elution. ODS column chromatography and RP-HPLC were also used. Five compoundswere identified as quercetin,astilbin,eurryphin,resveratrol,5-O-caffeoylshikimic acid byphysico-chemical property, spectrum analysis. The operational approach is simple, moreover, the elutingagent can be repeatly used. The activity inhibition anainst tyrosinase of five compounds were determined,the extraction with ethanol has higher activity than compounds. We supposed that the extraction withethanol has higher activity may be co-activated by different compounds or there was compound with strongactivity in extraction of ethanol, the mechanism deserved to be further researched.The resveratrol has pharmacologic action of antioxidant,anti-free radical and effecting the metabolismof arachidonic acid. The method which was used for quantitating the resveratrol of Smilax glabra byRP-HPLC was established,furthermore, the content of resveratrol from different areas were compared, thecontent of resveratrol from hunan xiangtan was the highest.The antioxidant activity of Smilax glabra was determined by DPPH·method, the results showed thatSmilax glabra which were collected from different areas has antioxidant activity, but the antioxidantactivity of Smilax glabra didn't has fixed relationship with the content, so we suggested that the mechanismof action was cooperation between different components.The total flavonoids of Smilax glabra from different areas were determined by spectrophotometri.The content was effected by the area and climate in some degree.
