Object:To detect the tissue expression of Angptl3 and LPL in brain,lung,heart,gastric wall,liver,kidney and adipose tissue of rat models with Hyperlipodemia,Diabetes Mellitus,IgA Nephropathy using Realtime PCR and explore the correlation of tissue expression level of Angptl3 with Plasma lipid,Diabetes Mellitus and IgA Nephropathy.Methods:The gene of GAPDG,Angptl3,LPL were amplified by reverse transcriptase polymerase chain reaction(RT-PCR)after total RNA being extracted from liver of SD rat and cloned into T-vector.The recombinant plasmids were screened and extracted to run Realtime PCR as templates.A total of 40 healthy male rats were divided into four groups:normal control(group NC,10),IgA Nephropathy(group IN,10),Hyperlipidemia(group HL,10),Diabetes Mellitu(sgroup DM,10).Group IN,HL,DM were fed with special food rich in lipid while group NC were fed with normal food.Group DM were injected intraperitoneally with small dose of streptozotocin(STZ) after two months.Group IN were injected with CCl4,lipopolysaccharide(LPS) and combining took orally immunogen bovine serum albumin(BSA).Using Realtime PCR(SYBR GreenⅠ) to detect mRNA expressions of GAPDH,Angptl3,LPL in brain,lung,heart,gastric wall,liver,kidney and adipose tissue.Results:The correlative coefficients of each reaction were 0.9976,0.9991 and 0.9996.The reaction efficiencies of each reaction were 0.9793,0.8623 and 0.8967.The Dissociation Curves showed only one kurtosis。Do well in modelling rat's HL,IN,DM which were similar with the clinical process. Gene of GAPDH was expressed in each tissue stably.Gene of Angptl3 was expressed in liver and kidney exclusively and expression levels were DM>HL>IN>NC。Gene of LPL was expressed in heat and adipose tissue mainly and expression levels were HL>IN>DM>NC。Conclusions:It was demonstrated that Angptl3 was expressed in liver and kidney exclusively,which would not chang with the levels of lipidemia,Diabetes Mellitus,IgA Nephropathy;There was a close relationship between the expression level of Angptl3 with TC levels,insulin-deficient,and IgA Nephropathy did not affect the expression level. There was a close relationship between the expression level of LPL with plasms level,Diabetes Mellitus。...
