| Objective:To investigate the expression of tissue factor (TF) in choroidalmelanoma (CM) cell line and in human choroidal melanoma, and to study itsspecificity that can provide some evidence on therapy of CM.Methods:1. CM cells (OCM-1) were cultured in vitro, and identified by observingthe morphous.2. CM cells (OCM-1) were cultured in vitro and TF expression wasstudied with immunohistochemical staining method in OCM-1.3. TF expression was studied with immunohistochemical stainingmethod in specimens with 10 cases of CM and 5 cases of human normalchoroidal tissue, and then analyzed its clinic significance.Results:1. OCM-1 growth curve was similar to "S" type, which included theadaptive phase in first 12h, then entered into logarithmic growth phase, andfinally into platform phase by 3d. 2. TF protein was mainly expressed in cytoplasm of OCM-1 (positivecell percentage 85.33%±5.47%).3. Among 10 human choroidal melanoma tissue, TF protein was mainlyexpressed in cytoplasm of CM cell cytoplasm (positive cell percentage41.60%±14.17%, IOD 33853.67±16445.30).4. Among 5 human normal choroidal tissue, TF protein was expressed inonly one case that mainly expressed in cytoplasm of vascular endothelial celland choroidal melanocyte(positive cell percentage 5.65%±4.76%, IOD426.43±316.62).Conclusion:The result suggested the positive expression of TF in both CM cell lineand human choroidal melanoma tissue, while negative expression or fakepositive expression in normal choroidal tissue which may play a role as aimmunotherapy target of CM. And it can provide some evidence on therapyof CM. |
