Objective: Hepatoma is the one of the most frequent malignant tumor in our country. Once they have been found, most of them have been in the intermediate stage or advanced stage. therefore many patients will miss operation chance. The development of interventional therapy make the life of hepatoma patients to get some prolong. But hepatoma is insensitivity to chemotherapy and easy to bring about drug tolerance, there are short of very effective therapeutic tool now for many hepatoma patients. Therefore looking for more effective medicine is still the focus of the hepatoma investigative by now. For the past few years, the research indicated that the acetylation and deacetylation of histone are correlated with tumor occurrence closely. Histone deacetylase inhibitor can obviously inhibit the proliferation of tumor cell. Although the mechanism of action still not very clearly, but it find out a new way for tumorous therapy. Now more investigations are applicating in those case, such as lung cancer, intestines cancer, bladder cancer, leukemia and so on. There are still no reports on the research of the effect and mechanis for histone deacetylase inhibitor TSA to the hepatoma cell SMMC-7721. For this reason, this experiment was designed to study the role of histone deacetylase inhibitors (TSA) to the hepatoma cell SMMC-7721 and its mechanism.Methods: 1. hepatoma cell growth curve were draw of experimental groups and control group through cell counting. 2. the hepatoma cells apoptosis was observed by Annexin V unite PI stain and Tunel experiment after TSA's action. 3. flow cytometry was used to study the influence of TSA to the hepatoma cell cycle. 4. Western were used to study the influence of TSA to the protein expression of hepatoma cell SMMC-7721.Results: 1. Histone deacetylase inhibitor (TSA) can inhibit the proliferation of hepatoma cell SMMC-7721 significantly. 2. TSA can induce the apoptosis of SMMC-7721 cells. 3. TSA can induce the cell cycle arrest at G0/G1 phase of SMMC-7721 cells.4. TSA can increase the expressions of p53, p21, bax and decrease the expression of bcl-2 in hepatoma SMMC-7721 cells.Conclusions: Histone deacetylase inhibitor TSA can inhibit the proliferation of hepatoma cell SMMC-7721 and induce the cell cycle arrest and apoptosis in the hepatoma cells. This maybe related to changes of expression of p53, p21, bax, bcl-2 and other tumor related genes.
