The Relationship Between The Serum HBVDNA Quantification And AICD Of Peripheral Blood Mononuclear Cells And The Expression Of FasL In Chronic Hepatitis B

Objective:To investigate the relationship between the serum HBVDNA quantifica- tion and activation-induced cell death(AICD)of peripheral blood mononuclear cells(PBMCs) and the expression of FasL in chronic hepatitis B.Methods: PBMCs were isolated from 30 patients with chronic hepatitis B and 13 health donors.The PBMCs were divided into two groups and were cultured for 72 hours in vitro.One group was added to the stimulation of PHA.The other was not added to PHA.After the cultured PBMCs with the stimulation of PHA were colored by PI,we began to use flow cytometry to assay the apoptotic rates.The others without the stimulation of PHA were marked by PE anti-human FasL,and then we used flow cytometry to detect the percent of the PBMCs whose expression of FasL was positive.Result: The apoptotic rate of the PBMCs in chronic hepatitis B was higher than the health group(t=4.93,P<0.01),and the expression of FasL was higher(t=4.65,P<0.01). The apoptotic rate of the PBMCs with different serum HBVDNA quantification in chronic hepatitis B was significantly different(H=18.0,P<0.05),and the expression of FasL were significantly different (H=10.99,P<0.05).The serum HBVDNA quanti- fication was high, The apoptotic rates of the PBMCs and the expression of FasL were also high in chronic hepatitis B. The apoptotic rates of the PBMCs had the correlation with the expression of FasL(rs=0.70,P<0.05).Conclusion: Activation-induced cell death in PBMCs was found in chronic hepatitis B.The expression of FasL in PBMCs was activated by chronic hepatitis B virus infection.The serum HBVDNA quantification was high,activation-induced cell death of the PBMCs and the expression of FasL were also high in chronic hepatitis B,and AICD had the correlation with the expression of FasL.
