Objective:To investigate the expression and significance of PTEN,BCL-2,P27 and KI67 in androgen-dependent and androgen-independent prostate cancer.Methods:The expression of PTEN,bcl-2,P27 and Ki-67 in 42 cases of prostate cancer (30 cases of ADPC,12 cases of AIPC) and 10 cases of benign prostatic hyperplasia were detected by immunohisto -chemical stain method and the relationship between the expression levels and pathological grades and clinical stages of prostate cancer were analyzed.Result:1. The positive rates of PTEN in benign prostatic hyperplasia and prostate cancer were 100% (10/10),40.5% (17/42) (P<0.01). The positive rates of PTEN in androgen dependent prostate cancer and androgen independent prostate cancer were 50%(15/30),25%(3/12),(P < 0.05). The loss of PTEN expression was related significantly to grade and stage of cancer ( P < 0.05 ).2. The positive rates of bcl-2 in benign prostatic hyperplasia and prostate cancer were 50%(5/10),42.9%(18/42) (P>0.05). The positive rates of bcl-2 in androgen dependent prostate cancer and androgen independent prostate cancer were25.0%(9/30),75.0% (9/12) (P < 0.05). Bcl-2 expression was related significantly to grade of cancer, but there was no signficantly relationship between Bcl-2 expression and stage of cancer.3. The positive rates of P27 in benign prostatic hyperplasia and prostate cancer were 60%(6/10), 45.2%(19/42)(P>0.05). The positive rates of PTEN in androgen dependent prostate cancer and androgen independent prostate cancer were 56.7%(17/30)16.7% (2/12),(P < 0.05). The P27 expression was inversely related significantly to grade and stage of cancer ( P < 0.05 ).4.The positive rates of Ki-67 in benign prostatic hyperplasia and prostate cancer were 20%(2/10),47.6 %(20/42) (P < 0.05). The positive rates of Ki-67 in androgen dependent prostate cancer and androgen independent prostate cancer were 36.7%(11/30),75%(9/12) (P < 0.05). Ki-67 expression was related significantly to grade, and stage of cancer.5. The PTEN expression was related to P27 of cancer.The PTEN expression was inversely correlated with BCL-2,Ki-67 in prostate cancer (P < 0.05)Conclusions:1.. PTEN expression was related significantly to grade and stage of cancer PTEN inactivation may play a role in progression to androgen independence.prostate cancer.2.Bcl-2 expression was related significantly to grade of cancer, the high expressionof Bcl-2 may play a role in progression to androgen independence. prostate cancer..3.The P27 expression was inversely related significantly to grade and stage of cancer. the low expressionof P27 may play a role in progression to androgen independence prostate cancer.4. Ki-67 expression was related significantly to grade, and stage of cancer. Ki-67 can act as a marker to the malignant degree and progress of the prostate cancer.5. PTEN,BCL-2,P27and Ki-67 have synergistic effect in progression to prostate cancer.
