Objective: To observe the relationship between the number of siderophage and wound time and their changing. To provide theoretical and practical evidence for the new histopathological method of wound age determination.Methods: The experiment was based on the model of mouse blunt instrument injury (laceration wound) and sharp instrument injury (incised wound) in wound area and the application of staining with Prussian Blue to detect siderphages on different interval after wounding was used. 145 Km mice were randomly divided into group and sharp instrument injury group .Each group has 70 mice and they were respectively divided into 14 different wounded groups according to different wound age(6h,12h,1d,2d,3d,4d,5d,6d,7d,8d,9d,11d,13d,15d),and every small group has 5 mice.5 intact mice were used as control. Then the mice were sacrificed and the sample was gotten for histopathological observation.Results :1 The Km mouse wound was observed to show obvious anatomical and histopathological change after blunt instrument injury and sharp instrument injury. Siderphages were negative in the controlled groups, whereas visible in the experimental groups, There was a remarkable difference between the experimental groups and the control groups in the number of siderophage(P﹤0.05).2.During the wound healing , the number of siderophages in wound area is correlated to the histopathology stage closely. The numbers of siderophage increased gradually with proceeding during the age of inflammation. The peaks were observed respectively at 7d and 5d in both groups, then the number of siderophage decreased gradually with proceeding.3.The injury in blunt instrument injury mice tissue was more serious than the sharp instrument injury's. The peak of siderophages number in the blunt instrument injury group's is higher than the sharp instrument injury group's and the time used to reach the peak is also longer than the latter.Conclusions: (1)During the healing,the number of MP in mouse insiced wound was correlated to the histopathology stage closely, and it was also correlated to wound age presenting typical bidirectional change. (2) The numbers of siderophage in wound tissue were related to the type and degree of injury closely.(3)The author inferred: the application of detecting the number of siderophage during mouse cutaneous wound healing by staining with Prussian Blue promises to be a new method for wound time determination.
