Background: One of anti-arrhythmia Chinese Traditional Medicine is WenXinKeLi which is widely used and its major component is GanSong (Nardostachy chinensis). The volatile oil of GanSong(VOGS) is utility concentration composition, and its detail pharmacological action mechanism is not clear yet.Objective: To investigate the electrophysiological mechanisms of VOGS by measuring the Effective Refractory Period (ERP) and value of corrected ERP(ERP/RR) of ventricular myocardium on rats.Methods: 52 Wistar rats were used and divided into control group (n=12) and treatment group (n=40) , 1:20K(3.4g/L) of VOGS was administrated . The treatment groups were subdivided into local infiltration group (n=20) and airway inhalation group (n=20). Electrocardiogram recorded the ERP of the ventricular myocardium. The ERP/RR as corrected value was used to evaluate the effect of ERP of the ventricular myocardium.Results:1. The effects of local epicardium infiltration group (LEIG) on ERP:1.1 In 15,30,45,60min, ERP is respectively 120.5±7.59,122.5±7.16,123.5±10.89,100.5±8.26ms,and respectively was compared with 66.5±4.89ms before giving VOGS and their relative control groups 65.0±8.37,66.67±5.16,65.0±5.48,68.33±4.08ms(p<0.01).1.2 The ERP, respectively ,was compared among 15,30,45min (p>0.05), but they, respectively, was compared with ERP of 60min(p<0.01).2 .The effects of airway inhalation group (AIG) on ERP:2.1 In 15,30,45,60min, ERP is respectively 119.0±8.52,127.00±10.30,128.00±11.05,97.00±12.18 ms, and respectively compare with 64.0±5.03ms before giving VOGS and their relative control groups 63.33±5.16,65.0±5.48,66.67±5.16,68.33±4.08ms (p<0.01).2.2 The ERP, respectively, was compared among 15,30,45min (p>0.05), but they, respectively, compared with ERP of 60min(p<0.01).3. The effects of LEIG and AIG on corrected value (ERP/RR) Comparing with a control group there are significance difference(p<0.01), which is similar with the effects of ERP.4. The Comparison of effects between the ERP with ERP/RRIn 15,30,45min, there are no significance difference in ERP between the LEIG and AIG (p>0.05), but there are significance difference in ERP/RR between the LEIG and AIG, because of adjusting by the heart rate(p<0.05). .Conclusion:1. The ERP of ventricular myocardium of rats can be remarkably prolonged by VOGS(1:20K)administrated through LEIG or AIG.2. The effects of ERP were prolonged and steady in 15-45min, then gradually diminished after 45min.3. ERP/RR adjusting by the heart rate can really demonstrate the effect of ERP of the myocardium of rats.4. It shows that administration strategy by AIG is better than by LEIG.
