Soybean saponin is one of the effective ingredients in soybeans, which is in the focus of food and nutritional research nowadays due to its numerous health-promoting properties. It is a kind of triterpenoidal saponin with many beneficial biological activities to human health. For example, promoting immunity, preventing cancer, lowering fat, lowering cholesterol and anti-oxidant activity etc. And it may apply in several fields such as drug, healthy food and cosmetics and so on.In order to study the physiological activities of relieving cough and resolving sputum of soybean saponin, the extraction, separation and detection method are studied. We used the soybean dregs as raw material, and made single factor extraction test to the ethanol concentration, extraction time and liquid feed ratio. Then we have carried on the orthogonal design to the main influence factor of extracting saponin from the soybean dregs, thus finds out the best arrangements of the three factors, namely Ethanol density 70%, time 12 hours and firm than 5 liquid. Through the color response and TLC, we made qualitative analysis to the extraction , and then made quantitative detection of soybean saponin by vanillin-perchlorate colorimetric method, witch is using the aglycones soybean saponin analogues oleanolic acid as a standard. By using the spectrometry method, we detected the total soybean saponin contents of the extraction samples we achieved in the experiment, and it is 19.3%. And the method is simple, reproducible and can be used to monitor soybean saponin content of the crude extracts.D101Macroporous Adsorptive Resins column was used to purify soybean saponin. By inspecting the eluting solvent, eluting solvent dosage and elution speed, we determine the optimum conditions for the elution are 100 ml70% ethanol solution elute following the rate of 0.4 ml / min. By using the spectrometry method, And we detected the total saponin contents of the purifying soybean saponin samples is 61.2% by vanillin-perchlorate colorimetric method, and the recovery of soybean saponin is 78%.On the basis of the above work, we confirm that soybean saponin has the activity of hemolysis, observing rabbit red blood cell suspension roled by soybean saponin, and minimum concentration of having the role of hemolysis is 0.6 mg / ml. In addition , through the mice which is irrigated saline, dextromethorphan hydrobromide oral solution and the mixed, we prompte them coughing with ammonia water. Then we record and analyze the cough incubation period, cough frequency within 3mins and trachea fluid. The results showed that both the antitussive and expectorant effect of soybean saponin are better than dextromethorphan hydrobromide oral solution, and the effect of the two mixed is no better distinctly than the single one. And the results of animal experiments do not demonstrate that soybean saponin has the hemolysis effects in diet.It provides a solid theoretical foundation, and has a strong practical significance for product development from this study on soybean saponin.
