Vaginal Delivery Of Mifeprostone: Effectiveness And Benefits

Background:Previously, mifepristone has been widely used in the clinics as an abortion drug . Since it induces resistance to the implantation of fertilized eggs, it has been also increasingly used in clinical applications as a contraceptive drug . Currently, mifepristone is mostly used through the oral administration . In this route, it is absorbed through the gastrointestinal tract and metabolized by the liver and kidney. Due to the severe side effects of the orally administrated mifepristone, it is banned to be used in people with certain diseases, particularly those with heart, liver and kidney dysfunctions. To avoid the "first pass" metabolism of the oral administration, to achieve better medication safety, and to expand the population of users, we explored new delivery channels for mifepristone.We used rabbits and rats as subjects for the vaginal delivery and the oral administration of mifepristone, respectively. At different times after the drug treatment, the physiological states and its contraceptive effects of the drug were investigated in the animals; the plasma concentrations of the drug, estrogen and progesterone were measured; and the changes in the tubal systolic functions were also examined.Objective :To investigate the administration of cholestero different way of animal state of the body and the research on contraceptive effect between two different administration MethodsMethodâ‘ Rabbits vaginal administration of cholestero(6ml/kg) and oral administration (6mg/kg) in different time, to measure serum concentrations of cholestero, serum estradiol and progesterone concentrations, tubal fluid, oviduct tissue Ca+ + concentrationâ‘¡Compare the damage effect on liver and kidney by vaginal administration with damage effect on the same organs by oral administration,and observe the slices of liver and kidney by electron microscopeâ‘¢compare the contraceptive effect of the two different administrationResultsâ‘ serum cholestero concentrations of oral administration is significantly lower than vaginal administration;Vaginal administration of cholester on serum estrogen and progesterone is significantly more than the impact on oral administration; Vaginal vaginal administration and oral administration is no significant difference in Ca2 + concentration and tubal smooth muscle contraction frequencyâ‘¡both the vaginal administration and the oral administration came to good contraceptive effect, contraceptive rate is 80%, 83%.â‘¢compared with the oral administration, the vagina were almost the same on the contraceptive effect,but in the electron microscope, there were certain liver and kidney damage of oral administration, no significant pathological changes of vagina administration. The serum drug level is higher if we administrated via vagina than others means. ConclusionSo little dose cholestero have the same effect and the same dose cholestero have better contraceptive effect, and have no harm to liver and kidney function .
