Immunomagnetic Microspheres (IMMS) are a kind of novel functional materials, which behave properties of magnetic materials and polymer materials. Because of their small particle size, large specific surface are, great capacity of coupling biology molecules, and good suspension in disperse system, IMMS are quite fit for application in biological system. Recently IMMS separation have exhibited extensive application prospect for their high efficiency,effectiveness,low toxicity and simple manipulation. This article briefly introduces the property,preparation and their application.In this experiment, IgG, which had specific bilirubin adsorption action with SPA, were separated from Porcine Serum by Magnetic Microspheres. The main content of this paper is such as following:Fe3O4 Magnetic Microspheres were prepared by means of chemical co-precipitation method and milling method. Such influencing factors as the agitation speed, the addition amount of surfactants, the milling speed, the milling time, the ratio of ball and material were surveyed. Some properties such as particle size, magnetic properties and iron content were characterized. The particle size of the product is smaller than 20nm with strong magnetic responsivity and superparamagnetism. The particle size distribution is homogeneous with good dispersion.Sepharose-SPA Immunomagnetic Microspheres were prepared by means of inverse suspension embedding method. Product structure, particle size and magnetic properties, were characterized and analyzed. Such influencing factors as the agitation speed, the surfactants, The indissoluble assistant were surveyed. The products' particle size is about 5μm, with good dispersion and exhibits superparamagnetism. SPA were coated to surface of Fe3O4 Magnetic Microspheres.The experimental results indicate that the best adsorption rate of SPA Immunomagnetic Microspheres is 70.12% and it can be repeatedly used for 29 times. This research provides some experimental foundations and bases for the industrialization and application of IgG separation from Porcine Serum.
