ObjectiveTo study the relation of HIF-1α's expression degree and apoptosis degree in cultured nucleus pulposus cell, and to study the relation of HIF-1α's expression and intervertebral disc degeneration.MethodsNucleus pulposus surgically removed from SD mouse of 30 days was used in primary monolayer culture. Then the cultured cells were cultured in different oxygen concentration to induce HIF-1α's expression in different degree. Western-blot was used to detect HIF-1α's expression. Flew cytometry was used to detect cell apoptosis degree. Correlation analyze was used to assessed correlation of the HIF-1α's expression and cell apoptosis in nucleus pulposus in date statistics of the results.ResultsThe four groups all expressed HIF-1α. The expression degree was different, and the difference is significant at 0.01 level. The four groups all presented cell apoptosis phenomenon. The apoptosis degree between four groups was different, and the difference is significant at 0.01 level. When the HIF-1αwas expressed high in groupB, the cell apoptosis degree was high. When the HIF-1αwas over-expressed in groupC, the cell apoptosis degree was higher than that in group B. When the expression of HIF-1αwas inhibited in group D, the cell apoptosis degree was lower than that in group B and C. The expression degree of HIF-1αand the cell apoptosis degree in four groups were correlated, and the correlation was significant at 0.001 level. ConclusionThe over-expression of HIF-1αis one of causative factors of nucleus pulposus cell apoptosis. And it was also deduced that excessive stimulation of hypoxia was an important causative factor of intervertebral disc degeneration and disc herniation.
